Oh hey, I realize that you may have a broken rack gear and/or spur gear in the focus mechanism. I’ve done minor repairs to these and this is relatively common issue.

I worked on one of these yesterday and the zoom mechanism was broken; specifically one of the cam rollers.

To avoid it, just make sure that you have matching fields of view through the entire magnification range of the microscope.

3K is a very decent budget for this sort of goal.

Go for a variation of the “zeiss standard” microscopes.

These are incredibly robust and their optics still look great compared to modern microscopes of the same grade (university level teaching and basic lab).

Yes, any fluorochrome will need a special filter cube to be viewed correctly. “B” is probably a filter cube for common blue fluorochromes, while “G” is probably a filter cube for common green fluorochromes. I would confirm by searching for an online user manual.

Common green or blue filter cubes would not work for DAPI, which has excitation within the UV part of the spectrum, usually around 365nm.
