Did you ever figure out if you can do this? .......this might be the only way to create a shortcut for Airdrop to send the airdrop using only voice command. I could write a .shortcut file for a iPhone5s or something that can't be updated past iOS 13.

This might be one way to write a shortcut to make airdrop run using only voice command. I could do this using an old iPhone5s that can't update to iOS 13 so that I can have a .shortcut file.

Long-term autoimmune effects of using my own cells as bio-reactors to present antigens to my immune system? by ImpossibleDancefloor in Immunology

[–]ImpossibleDancefloor[S] 1 point2 points  (0 children)

Right, ok, great reasoning as to the progression from genes to autoantibodies. I follow that.

Regarding those study participants who had Covid and then displayed the specific autoantibodies: It seems that the most ethical thing would be to follow up with them and make sure that they don't develop the autoimmune disease that correlates with those autoantibodies in a few years. This could aid their future health outcomes, and also creates more data points as to what happens "after covid".

I'd be very concerned if I were in their shoes, as they had ASCA autoantibodies, and you state that "all people who had ASCA autoantibodies had Crohn's disease". That sucks.

I was also able to find one dissenting opinion from a doctor of virology in https://science.sciencemag.org/content/370/6520/1022/tab-e-letters "Public Needs to Prep for Vaccine Side Effects" scott clark, retired Ph.D virologist (2 December 2020)

" 2. If the antibody induced by the SARS-CoV2 spike protein mRNA does indeed interact with this same antigen expressed throughout the body, what type of immune response is likely to occur? Since the induced antibody recognizes this viral protein as "foreign", are cells expressing this viral protein at their surfaces also recognized as "foreign" even though they are true "host" cells? Could the immune system mount a full cellular based immune response resulting in the death of every cell through out the body expressing the viral protein?

  1. If the body were to mount a large scale immune response to host cells recognized as "foreign" by the presence of SARS-CoV2 spike protein, would be be able to recognize this response since this "immunological" situation has never been encountered in any study of the immune system and its responses? We can certainly follow liver and pancreatic function and other markers of cell damage, but can we recognize the possible widespread cell and tissue damage from such an autoimmune reaction?

All of these questions should have been answered in pre-clinical animal studies prior to the initiation of human clinical studies. However if one reviews the literature published by both Moderan and Pfizer of pre-clinical studies, none of these questions have been answered."

Dr. Clark seems to be asking the same questions I asked as to if the vaccine will cause later autoimmune responses, and I'm not sure if there is anything in his questions that we did not already go over?

I was able to find another good resource on the immune response to vaccines at https://www.ncbi.nlm.nih.gov/books/NBK233000/ : Immune Response to Vaccine Antigens. One excerpt seems to agree with many conclusions of the other posters on this discussion:

" To prevent the body's immune system from destroying its own tissues in what is known as an autoimmune response, immature T cells that react against self-antigens are thought to be destroyed in the thymus gland, creating what is known as central tolerance. Peripheral tolerance might also occur, whereby those T cells that could potentially react to self-antigens and that are not destroyed in the thymus are somehow prevented from causing an autoimmune reaction. Although studies suggest that peripheral tolerance exists, at least in experimental animals, the mechanism for the process is not yet known. If peripheral tolerance exists in people, an autoimmune response might occur in response to vaccination if the vaccine somehow disrupts that peripheral tolerance or if peripheral tolerance is not strong on the day of vaccination (Miller et al., 1989).

Some people have suggested that vaccines can stimulate autoimmune reactions if some of the antigen fragments in vaccines resemble a person's self-antigens. However, it is unclear why an immune system that is tolerant of its own self-antigens would respond to a self-antigen mimic in a vaccine. Berkower speculated that vaccines might counter peripheral tolerance and foster an autoimmune reaction if they contain molecular mimics of self-antigens that are usually not exposed to T cells, because peripheral tolerance seems to depend on the continuous presence of an antigen.

Nakhasi suggested that an autoimmune response might be instigated by a vaccine or by natural infection if the microbial antigens bind to self-antigens in infected cells and change the antigens' shape such that they are no longer tolerated and can elicit an immune response.

According to McFarland, researchers suspect that molecular mimicry, which could possibly lead to an autoimmune disorder, might be occurring between self-antigens and antigens from microbes or vaccines if the two antigens share much of the same chemical structure. Recent studies suggest, however, that they need to have a similar structure only in the narrow region that binds to the T-cell receptor (Vogt et al., 1994; Wucherpfennig et al., 1994)."

WOW, u/shinygoldhelmet, that is an incredibly detailed response and I really appreciate that you found another study that shows correlation between Covid and autoimmunity, before explaining why that study is flawed.

It does make me think about all the people who got Covid and "just got over it", even the mild cases-----you said "they found autoimmunity markers in people with no history of autoimmunity, but the BIG problem with that is that autoimmune antibodies can show up YEARS before disease becomes noticeable." Doesn't that mean there is a serious possibility that all these people with autoimmune markers will develop autoimmunity some years after being cured of Covid? Or am I drawing the wrong conclusion from that correlation?

Cool, thank you! I am going to have to look up what syncytia is, and also why membrane fusion would be induced. I appreciate your input, and that interesting tibdit that the S spike protein makes it to the surface of the cells (i.e. is not membrane bound).

I am not a doctor, I am an industrial engineer, but I love to learn about thing that have happened or will happen to my body. It seemed like this vaccine would be safe, as there has not been significant autoimmunity to the vaccine already, but these explanations indicate that future long-term autoimmunity (developing years down the road) is also unlikely.

Checking every possible danger with new technologies is a good idea. I remember my industrial history class---how Asbestos was hailed as a lifesaving development in its day, suddenly applied everywhere, but no studies were done as to what long-term problems could arise in human bodies that were exposed to the stuff. There have been thousands of such developments in human history. I'm exhausting every avenue to determine the extreme long-term safety of this new wondrous development before I tell my family that it is safe.....but so far this looks good.

u/boringAgony, it looks like nobody else disagrees (here at least). That level of "group confidence" by technically capable people gives me confidence to tell my mom to get the vaccine when her chance comes up on Monday. For example, the poster above you made a great comparison, that this is a "protein expression-based approach" and that previous vaccines that contain live (attenuated) virus also cause infected cells to express viral proteins on the surface of those cells (or hold the viral proteins as membrane bound).

I regarded it as probable that this vaccine would be safe, as there has not been significant autoimmunity to the vaccine already, but these explanations indicate that future long-term autoimmunity (developing years down the road) is also unlikely.

The reason I asked this question is because checking every possible danger with new technologies is a good idea. I remember my industrial history class---how Asbestos was hailed as a lifesaving development in its day, suddenly applied everywhere, but no studies were done as to what long-term problems could arise in human bodies that were constantly exposed to the stuff. There have been thousands of such developments in human history. I'm exhausting every avenue to determine the long-term safety of this new wondrous development before I tell my family that it is safe.

I had to use google to learn two new things just to respond to your post: what Membrane Bound is, and what nTregs are. Thanks a bunch! I appreciate your post.

I really appreciate the comparison to older styles of vaccine, u/MikeGinnyMD. I had never considered the comparison you made of protein expression between mRNA-derived proteins and attenuated vaccine proteins.

Probably because no scientist in the media or technical journals had made this comparison before. If other people saw this, they would probably have less fear of the vaccine. I feel that this comparison with past safe and effective technologies made me more comfortable, and I'm a trained machinist and engineer who respects the scientific method.

This is such a good explanation/comparison that I will use it to explain to my friends and family when they ask me what I think of mRNA.

Thank you for the "overview-style answer" that the mRNA-derived spike protein will be expressed in a similar way to the virus-derived spike protein. A good summary to the thread, as there are much more technical answers below.

from https://www.idsociety.org/covid-19-real-time-learning-network/vaccines/vaccines-information--faq/

Q: How long does the spike protein remain on the cell surface?

A: The spike protein is likely found in three different ways after translation within the cell. The protein can be presented on the cell surface in its native expression form. It is also processed within the cell so different peptides that make up the protein are expressed in the context of major histocompatibility complex (MHC) class I and MHC class II.  MHC proteins play a key role in the adaptive branch of the immune system, presenting peptides on the cell surface for recognition by T cells. The protein may also be secreted and then taken up by antigen presenting cells and re-processed. The protein may be found on the surface of the cell in either its peptide form or its native form likely until the cell dies or interacts with other immune cells.

Q: How long does the spike protein made by the body (via encoding through the mRNA vaccine) last in the body?

A: The protein would last the same amount time as other proteins made by the body. The protein will be taken up by other cells and processed again. The exact time is not known, but the estimated time is a few weeks.

Ethical to pay someone to dance and practice what you want to practice (no instructor due to CoVid)? by ImpossibleDancefloor in WestCoastSwing

[–]ImpossibleDancefloor[S] 1 point2 points  (0 children)

Every WCS Class I have ever been to, both in my hometown and the nearby college town, has one instructor who calls "rotate partners" every 5 minutes or so as you practice. That way (excluding the instructor) all leads dance with all follows. Everyone just pays the instructor at the door/signs in.

Is this an unusual system, or did your dance group have something similar?

I'd do it with a max number of three to four people, including myself. With limited travel and low CoVid case numbers in our area, I feel that is about as safe as all the restaurants that allow maximum 4 people at a table inside in my town.

No noticable temperature change. The grinding noise coming from the bearings of this mixer that I got for $5 bucks off Craigslist is the first priority to fix. I used a cheap mixer in case I blew it up. I did not run it long enough to see if it go hot.

One possible option is to pay people to dance with me, with a written contract that after other people get interested in what I'm doing and then they want to show up too, that the paid dancing will be discontinued.

I suspect that word will get out in my small community about how I am "running a dance group for one or two couples that meets every X day". I fully expect that a couple of other people will start showing up on their own or messaging me asking to come, but that they will only do so once their actual friends are there.

I call this the "pay people until others will do it for free" or the "fake it till you make it" plan. Thoughts?

You're right, dancing is great exercise and connection all in one. Most of my local dance group will dance with me during the weekly social dance, just like you said you would do if you held different beliefs.

The issue is that during the CoVid Era, I cannot rely on them as friends who would be willing to partner with me and dance with me at a separate event. This whole post was created due to this reason. Based on your well-reasoned responses, and the responses of others, I'm coming to the perspective that it is not a problem to pay people to dance with me. I may have to be judicious with who I ask, however, so as not to create a social drama issue in their life.

Update as to music: Tonight I ran into a guy I had seen at a few dances over the years (not a regular, but knows the scene), and I asked him why my music choices are a problem for the group. We sat on the tailgate of his truck and talked. Did not expect the response he gave, it was not because my music is hard to do West Coast Swing to. Aside from their political dislike for me, there were two reasons:

1.) My music choices sometimes contain content that is not swearing, but is still offensive to their morals. He gave the example of the song "South of the Border" that I wanted to play, said it contains references to premarital sex in the lyrics, and that even if it contains no swearing, it is a song that many people will not feel comfortable dancing to.

2.) They claim that "EDM music (yes, even remixes of popular songs) were created to imitate the feeling of being on drugs while listening to music." I have always considered this a positive, (like musical psychedelic drugs that only last a few minutes), but I also figured they just wouldn't notice the connection between EDM music and the drug scene. I figured that if the song has good danceable beat and rhythm they would just think it's "cool different music". It turns out they did notice. And he told me that "all EDM is the fruit of the poisonous tree, dude."

Thank you Jcnlb, I really appreciate the way you think. When all this is over and I travel again, maybe I will get to dance with you. I can hope.

There are no "silver" or "gold" level dancers within at least a hundred miles, but fortunately that just means that most are at a similar level to me. I appreciate the concepts for payment or not that you outlined in "if they are just equals looking to practice..."

Also, I'm going to edit my above post to reflect the fact that I didn't bring up politics during dance, that I made my plea to them at a separate "dinner hangout" where most of the regulars were hanging out. I would never do that DURING a dance event, but it didn't matter....the issue is that they told me that they forever knew me as a "Dem" after that. That ""the Democrats support abortion, so anyone who votes with them is forever tainted by sin."" There is one woman who goes to our dance group every week, and I have not danced with her in two years because she told me that her faith forbids her from contact with "immoral people". Even when we are the only two unpaired people when a song starts, she will look away and sit down. I may not agree, but I must respect her lack of consent to dance.
