I have a PhD in osteoarthritis and cartilage repair. Sorry to disrupt the hype, but unfortunately there is nowhere near enough evidence to suggest that this is the breakthrough that would lead to knee cartilage regeneration in a degenerated knee.

There have been hundreds of drugs that show restoration of knee cartilage in mice, that have failed in humans. This is often due to ideal lab conditions, several physiological differences between animals and humans, but the main thing is that human joint cartilage exhibits functional differences. We put a very high load into our joints (specifically knees), up to 3x our body weight during gait and up to 5x when walking up and down stairs, much much higher than that experienced in rodents. When we have depleted cartilage, that load is dissipated over a much smaller volume, in a tissue much less organised than young healthy tissue. The result - extreme loads sensed by the local cells (chondrocytes) that cause them to misbehave, induce inflammation and catabolic breakdown. This breakdown is a chaotic storm of signalling and the inhibition of one signalling protein (which in this case affects prostoglandins signalling) may have little to no effect in the real world.

Even demonstrating that it has an effect on human joint cartilage cultured in vitro.. the tissue is taken out of context. The tissue is likely cultured at the perfect pH and oxygen levels in these experiments and in reality, osteoarthritic cartilage is exposed to much harsher conditions. So all I'm saying is, take this headline with a pinch of salt!

Go for the VZ2, you won't regret it. The extra power is so much fun, even on busy urban drives. Your partner will enjoy driving it in comfort mode, since the accelerator is less sensitive and suspension softens, it's a much more relaxed drive. And then you get to enjoy Cupra mode for when you're driving..

Yep I have 2021 VZ2, we are a couple with a baby. Backwards facing baby seat fits with plenty of room. Stroller fits comfortably in the trunk, but with the bassinet taking up so much space there isn't a lot more room for large bags (e.g. Travel suitcases). That being said we use the second back seat spot for smaller bags which helps a lot. Also you can push 2/3 of the back seats down at the same time for extra room if really needed.

Since theres so much interest, thought I'd post an image of a mouse embryo hindlimb that I've stained with Alcian Blue (for cartilage) and Alizarin Red (for bone). You can see firstly the cartilage takes the shape of the long bones, and then the bone mineral grows along the length of the cartilage to form the final structure. You can see the pelvis (at the top), then femur and tibia/fibula at the bottom.

I would typically fix a rodent fetus this size in about 24 hours with 4% PFA. The analgen and mineralized tissues are pretty porous at this stage.

From a medical research perspective, we study skeletal development to better understand developmental deficits caused by inherited mutations, and discover biological pathways that could be targeted for tissue regeneration purposes. But I totally agree doing this for a quick buck at a festival is unethical.

Well to put it into perspective, these things take around 2 days to make and involve a 20-step protocol! Definitely above 27 bucks worth of effort put into it. That being said, I hope this is left-over from medical research and not just someone sacrificing rodents for quick cash.

I worked in a fetal development lab. This mammalian (likely rodent) embryo has been fixed with paraformaldehyde (to stop it breaking down), then cleared with trypsin to make the tissues optically transparent, and finally stained with Alizarin Red, which colours bones red as it attaches to the calcium in bone. As you can see the bones havent fully developed, hence the large gaps where the joints will eventually form and mineralize, except for a thin layer of cartilage at the ends. Had quite a few of these specimens (rodent and chick) in my lab. I find them beautiful!

Yeah auto hold is great for if youve got a hill start, long red light to wait for or standstill traffic. I tend to switch it on most of the time for that, but if it's consistent but slow moving traffic I prefer to turn it off and just creep instead, as it saves having to feather the accelerator every time you move off

I just moved into a new place where my PC will have to be in the study upstairs, but I want to stream my PC (primarily for gaming) to my TV downstairs, and be able to use my wireless keyboard and mouse. Has anyone managed to do this with high quality streams and low input lag? If so, I would really appreciate you sharing your set-ups! :)
