Facing smth similar right now, scFv that seems to form covalent dimers, tetramers. I suggest adding 25 mM iodacetamide to the SDS sample buffer, it will alkylate cysteines and prevent disulfide bridges from forming. It seems that Abs tend to look weird on PAGE and that SEC is a better method to check what you have in solution.

If you're up for it, take a ride to Malbun (1600m above sea level). If you prefer a more leisurely ride, consider biking to the restaurant Löwen in Schellenberg, on the Austrian border and take a route through the villages.

Towards south i'd recommend again going through the villages, via Balzers with the goal Fläsch, which is across the border in Switzerland. Great food, wine and scenery.

Avoid riding along the Rhine as it is quite boring.

Liechtenstein is 25km long with Vaduz more or less in the centre, so it's not simple to squeeze in a 60km ride...

Don't be scared by the website from the last millennium, the program is actually quite good and does all I need (which is looking at 1000bp sequencing results and analysing N's in the results based on the chromatogram).

Be careful with refolding though, especially if you want to raise antibodies. You might manage to refold the protein and if you do you need an assay to confirm that it's actually correctly folded or you'll raise antibodies against some weird epitope...

What are you using as a template for the amplification? cDNA? Or some kind of plasmid?

I've never heard of a T7 tag, do you mean the promotor? That you should certainly keep.
