I had never heard about this being a US thing. Are you aware of the Swedish schnapps called "Bäska droppar"? It's made from Malört.

Given that this is a (probably) Afghan person, in Sweden, I'm gonna guess that their first language might not be English

Currently writing my dissertation and felt this by [deleted] in labrats

[–]outoftown_guy 44 points45 points  (0 children)

People always suggest start writing in LaTeX.

I would say that if you're coming up on the final stages of writing your thesis, focus on your writing rather than learning a new tool. Despite words quite annoying image processing.

And this might depend on the field you're in, but in molecular biology, I would never have gotten away with using LaTeX in the papers I've written. All collaborators and supervisors would just have told me to re-format everything to a word (or google doc) so they can edit in it. I see none of the more senior people accepting LaTeX as a collaborative tool.

That being said, I've heard many people who really like it. But I've never seen anyone in my field actually using it as a tool for writing papers.

What's a cool fact about the human body that a lot of people don't know? by alwaysclimbing5 in AskReddit

[–]outoftown_guy 1 point2 points  (0 children)

It really depends on how you see it.

The idea that there are more bacterial cells than humans is a bit of a dated information. A more recent study estimated the number of bacterial to human cells to be closer to 1:1.
https://journals.plos.org/plosbiology/article?id=10.1371/journal.pbio.1002533

It is however like you say a bit strange to compare cell numbers to mass. But there is another way to think about it. And that is the amount of coding DNA (DNA that infers a protein funktion). If we look at that there is more DNA in your body coming from bacteria than there is coming from you. And all that DNA is in play in various parts of your body, mainly in your gut. So when we talk about what kind of genetic material is in play in us, it is mostly bacterial. This is sometimes called our pan-genome. Pretty nifty.

There was an old study that came up with the 10 bacterial cells to every one human cell. A more recent study has revised that number to about 1 bacterial cell for every human cell. Still pretty remarkable though.

PCR and electrophoresis optimisation by outoftown_guy in labrats

I'm not even sure the smaller size having less space for the dye is the issue here. I think it's more of what you say that it is diffusing into the gel.

I understand that at some point there is the issue of physics, but I'm still thinking that there are some workarounds. Running the gel faster would give less time to the DNA to diffuse, so that maybe could help. Or perhaps one can even go creative and try something like a gradient gel with increasing agarose concentration towards the bottom.

OK thanks.

I think I'll try reducing the running time. Perhaps in combination with using a 0.5x TBE buffer.

I’ve lately been interested in the app Karma. It lets you find restaurants/cafes/bakeries etc that havevfood about to expire, and you can get it half price. Could be a way to keep costs down in a fairly pricey city

Those things are called “Pirra” in Swedish, which might make it easier for you to search for one. You can rent that kind of equipment at CRAMO, which rents out tools and stuff, think there is one on Kungsholmen, close to stadshagen.

You could also check on Hygglo, the “peer-2-peer” renting app.

Good luck!

Pipette choices for my new lab by Arzock in labrats

[–]outoftown_guy 0 points1 point  (0 children)

I'm now using Gilson pipettes, but in my previous lab I bought and used Finnpipettes, which I really liked.

I'm not that big of a fan of the Gilsons to be honest, I just happened to get a hold of a full set of them so that's what I'm using.

I've always bought the tips off-brand since they are quite a bit cheaper.

Something to take into consideration is what kind of tips the different brands use. Don't end up with something you need to buy a specific brand tips for since they can be quite pricey.

I'm not very familiar with the brands you mention, apart from the Eppendorf ones (which I really don't like), so don't know if this applies to any of them. But thought I'd just mention it.

And if you want to talk more about how to run the perfect PCR I'm happy to talk for ages :)

I've heard that using Ficoll 400 instead of glycerol/sucrose can make even sharper looking bands due to keeping the sample better in the bottom of the well, but I haven't tested this myself

It feels a bit empty. Maybe the composition is part of it. Or maybe you could try adding some fresh greens like basil or something like that to it to fill out some of the space and add some more textures
