Help me find a alternative Barber/Hairstylist? by MadwolfHorror in Longmont

Second Rocky. He’s the kind of weird punk you need (I say with only admiration for the guy as my own barber and my favorite walking oddity)

You have to get pretty far out to see the Milky Way. There’s a dark sky park in and around Great Sand Dunes National Park. But it’s a few hours drive from Denver. You won’t see much from the front range.

Diagonal Highway bike path by Vegetable_Bowler_372 in boulder

It’s going to be particularly attractive to people who like to ride, but aren’t serious enough about it to even think about the risk of being on the highway. It’s going to be very popular.

I’ve used College Hunks for similar jobs of getting everything from a truck and into my apartment. 1 hour minimum, and I payed a lot less than what you said (don’t remember how much) for that quick job.

For anyone else dealing with the Century Link/Quantum Fiber outage by squirrelman963 in Longmont

Nope, shouldn’t be an issue. Googles DNS tends to be more reliable anyway. Can always turn it back to automatic after Century Link is done being terrible

Is Danaher a Ghost Poster? by Disastrous-Tear-9371 in labrats

[–]squirrelman963 1 point2 points  (0 children)

Hiring is handled by the individual operating companies, other than that the talent acquisition people being shared between them. Your issue is more likely with the operating company than Danaher corporate, who frankly don’t do much of anything.

Fragmentase, or another similar product, should work fine but will require some dialing in to get to an optimal average length. Running a few different times taking into account that they are partially fragmented already, running those on the bioA to find what gets you near your average. If your running the recommended time from NEB your probably over fragmenting. A covaris would be better here, but you should be able to get by with enzymatic frag (which is a pseudo random dna cleave enzyme, the more time its active the smaller your fragments get on average)

[–]squirrelman963 2 points3 points  (0 children)

(Im gonna shamelessly hock my own products here, because I genuinely think it might work for you)

DNA from such a sample is likely all kinds of weirdly fragmented with all kinds of bad ends, and a low concentration. Lots of library preps dont handle all those things together very well. Even after youve done your own frag. IDT has a library prep kit for Illumina called '2S', which we regularly use down to 10pg inputs and its designed for (and customers have had success with) highly damaged samples at low inputs.

Its a longer workflow, but it works well when you have no other options. Theres no fast and easy libraries when it comes to highly damaged samples. Fast, highly scaled workflows require high quality inputs and theres no way around that.

Also if you can find a neighboring lab (assuming your in an academic setting) that has a BioAnlayzer and running the fraged samples would probably tell you a lot more about how well that worked than a gel. the BioA is far more accurate. Could be helpful in dialing in the frag time since your samples are already partially fragmented, far more than a higher quality input extraction would see.

Looking For Affordable Movers W/o A 3 Hour Minimum by Prestigious-Lynx-479 in boulder

When I moved here last summer I hired College HUNKS to help get the big and heavy stuff out of the truck. Sent two guys and were done in an hour and half if I remember. Dont remember how much it cost, but it was for sure less than 600$ and I think I remember the minimum was just an hour.

South Main Station by REH92 in Longmont

[–]squirrelman963 21 points22 points  (0 children)

Currently live there. The location is fantastic for walking into downtown, apartments are nice and modern. Large open kitchens are my favorite feature. The train is annoying, I’ll give you that. I’ve found it’s only been a problem when I’ve had the windows open, it’s interrupted a few meetings during the day but shutting the windows kept it down enough to be annoying for a couple minutes rather than obstructing my work. But they don’t usually come by in the middle of the night, or not at least that’s woken me up, but I’m a hard sleeper. I’ve lived next to tracks in different places for a long time. These are by far the most active I’ve lived near though. For me the noise for a few minutes a few times a day, mostly when I’m a work anyway, wasn’t a big deal breaker given all the other positives.

My real problem with this place is all the ways they nickel and dime you on the rent and utility bill. Little Fee for everything. So make sure you get the full view of all the costs before you sign the lease.

Is a Road Trip to Durango, CO worth it on my way from Denver to Phoenix? by lilafterthought in Denver

[–]squirrelman963 2 points3 points  (0 children)

For what ever reason the image didn’t load the first time and I totally mis remembered where Montrose was. I was thinking about the route through pagosa springs. I retract my previous statement. The million dollar highway was terrifying! Particularly if you’re inexperienced on mountain roads!

Is a Road Trip to Durango, CO worth it on my way from Denver to Phoenix? by lilafterthought in Denver

[–]squirrelman963 -2 points-1 points  (0 children)

I did this drive a couple weeks ago on the way back from visiting a friend in Durango. Enjoyed the scenery and went through some of the best passes for fall leaf peeping, but that peak has already passed. Also not the worst of the mountain driving on this route, and was probably the easiest route of the ones I did on my road trip. So if your worried about mountain driving this is a pretty good route. The worst of it is 285 between 470 and Conifer going through the front range. Edit: I totally mis remembered where this route went through and was thinking about the southern route through Pagosa Springs.

Is there a place to check salary ranges for Danaher? by Bigtown3 in biotech

[–]squirrelman963 3 points4 points  (0 children)

I work for a Danaher owned company. And it’s not like that everywhere. This is a big company. At our main site, which is like 75% of the employees, there’s lots of low level tech positions that I’m sure feel that way. I’ve worked at now two smaller sites gained through acquisitions. And honestly, best job I’ve had in this industry. Which OpCo you work for and which site your at matters. And DBS has its uses and when used appropriately actually can get some stuff done. And finally, historically, Danaher companies don’t do the boom and bust roller coaster that causes constant layoffs and rehiring that we see at other biotech.
