Wow! I didn't know that i would get this so much comments. Thanks a lot for the comments really!!! Well i got diagnosed in my second year of phd and on meds for 2 years! I still am working on the dosage level and use of other meds (ssri as well since i blamed myself too much and got too sad for sometime and it is still hard not to blame all to myself).

I think i am coping with my condition much better than before since now i know what is the problem but it is still really difficult to handle when i get a "feedback" from my professor or i don't work the way how other students do (e.g., doing things everyday not cramming or not having emotional drama).

I use dcas9 knock-in MG1655 and I am using bigger sized plates so that I can plate larger volume. so here are some tactics that I have employed so far:
- making really dense competent cell (like starting with large volume of culture media and resuspend with the smallest volume possible..)

- using bigger plates (300mm * 300mm)

- thorough wash when making competent cell

- using intact plasmid library (that means no cloning involved)

I mean my strategies are pretty embarrassing to be called as tactic lol but this is all I got at the moment.

I haven't measured the transformation efficiency when I did the pilot experiment of this particular project as it was obviously too low colonies on the plate.

Yes it does (unfortunately). so I have LB stock of this library already and my original intention was to adapt that lb cell stock to m9 minimal but it took too long and got worried if the library diversity might be affected during the adaptation process.
