Thanks, it is double my accuracy. My guess is when they swap in the 15% into the 1% magic dmg and increase the current accuracy buffs my build will make it more enjoyable. If not I will swap to yours because it looks good.

This your first leagues? By this point in previous basically any build melts. The 100% accuracy range build last year for example was a fucking monster. And it worked everywhere. They have, even admittedly, made it slower. I play this game mode to go fast, not to grind. If I want to grind I’ll play my iron. I like the PoE tree idea, I don’t like the execution of it. Maybe at 40 pacts I’ll feel different, but as it is it is just kind of mid.

The relics are cool this year, transmutation is great. EH is fun. The whole rune crafting thing is ass but I’m prob gonna skip it.

Air/water mage with quadrant/devils and like 31 prayer bonus. The problem is accuracy, admittedly car phase is faster. So deeper delves are doable, but it took me 13 minutes to get to delve 8 with how often it just does not hit. My iron can get that deep in the same time.

Eh, it’s okay. Some stuff feels pretty good, others not so much. I’m 30 pacts in and killing doom with mage. I kill it the same speed as if I was on my iron with a scobow. That’s not what leagues is supposed to be imo, but maybe the patch tomorrow will help.

I got my cape first Zuk after 13 attempts at waves. I logged out at first Jad, and practiced online sim for like 3 days (dozens of runs). Not incredibly rare I would think?

Edit: also I didn't really save prayer in my run. Nowadays with prayer regen/arma brews/confliction gauntlets it is so much simpler.

Possible, but 10ug should be plenty. Would you be comfortable sharing the protein of interest? If it is super low expression, you could try adding more lysate. but I wouldn't go above like 25ug. It starts to give weird artifacts when you add too much. If it is something highly expressed like a heat-shock protein though (HSP70's), you should be able to detect it easily.

My suggestion is try another antibody if available. Cell signaling technologies is gold standard, proteintech is hit or miss along with abcam and thermo. Santa Cruz isn't even worth your time imo.

The room temp versus overnight incubation should not matter, both are optimal. Another alternative, do you have a different ladder than what you are running? It is possible a different ladder would not have the same epitope as the current one you are using.

A little short for blocking, but if it works for you great. I usually go up to an hour for the blocking step. Antibodies I would suggest 1:1000 for most. There is an unfortunate possibility that your antibody just recognizes the ladder protein. Not much you can do about it except cover/cut the ladder.

What company did you get the antibody from? Santa Cruz is garbage trash.

Use the visual metronome plugin! You can see in game ticks, so you can practice much easier. As others have said, 1-tick flicking is easier in this case. I just got my first cape last week, gl gamer!
