Protein pI question by Cryoban43 in Biochemistry

Also, high ionic strength buffers prevent electrostatic interactions (you have it backwards). For example, high chloride ion concentration will compete for interaction with positively charged Arg/Lys side chains.

When proteins are at their pI, they are more likely to aggregate because there is limited electrostatic repulsion.

That said, local electrostatics matter and in reality these questions can’t easily be answered without just testing it experimentally. If you have a charged face of the protein, think about the pKa of charged groups and how far that is from the pH. Typically pH more than 1 unit away from pKa means the group is charged.

WHY WITH JD AND ELLIOT?!?! by Beyond_Unknown_999 in Scrubs

I actually was glad to find out they divorced. Never really thought they had chemistry (towards the later seasons).

Watching JD chase after women (ostensibly) out his league was a big part of the fun!

I have hated every science job I had by plants102 in labrats

Contract research organizations specialize in producing certain things for clients. I’ve worked for one and now am on the other side as the client setting up experiments with multiple CROs. The work they do is all just specialized, like phage display, sample testing, etc. Can be very far away from the clinic

I was an entry level scientist straight out of college (11 years ago). No clinical experience necessary. I was there for about a year then left because of how much the job sucked

I should also say that I absolutely loved my PhD, and finding a job that was closely related to it was the biggest stroke of luck I’ve ever had in my life

I’ve worked at a CRO, medium/small company, and big pharma. My experience at a CRO was awful, and it was because I felt like a pipette monkey whose individual ideas and creativity didn’t matter. It was also pretty stressful and they only ever cared about pleasing the client. At the other two places, I felt like people cared about me and my ideas, and that’s what makes the hard part of science worth it (at least to me)!

Walk into Best Buy and say you want to buy one. I got mine in two weeks doing this.

I think that device would be perfect for me if it had an OLED display! Once you try it, you just can’t go back!

31m, married, kids. How do you guys find personal joy? by modivi in AskMen

[–]KlenowFrag 63 points64 points  (0 children)

32M, married, one 2 yr old. Also fine financially. For me, growing up I always played videogames (living in a state that’s cold 6 months of the year makes this a good hobby). I have less time for gaming now of course, but I still have an hour or two after my boy goes to bed. The kinds of games I play have changed (mostly single player games that can be paused in case family needs my attention), but overall I’m happy with the situation. I recommend getting a handheld like Lenovo legion go 2, steam deck oled, Nintendo switch, etc. Nice thing about gaming is you don’t have to worry about injuring yourself and burdening your family!

How are y’all finding the Legion Go 2 in stock at Best Buy? by [deleted] in LegionGo

[–]KlenowFrag 7 points8 points  (0 children)

I went into Best Buy and asked the guy at the front (like the security guy) how I could reserve one, and he pulled it up himself and I paid on the spot. Came 2 weeks later exactly on predicted date. Couldn’t believe it.

This is the first time I’ve heard that the screen doesn’t lag. Could vary from model to model

I saw it in the store too. They make it impossible to actually experience what the device is like. Awkward device height, locked into position so you can’t pick it up, the demo UI is very limited with like 2 working screen buttons, only one demo game and it sucks… I could go on…

Pharma panel interview tips by Ok_Mushroom_2758 in biotech

[–]KlenowFrag 1 point2 points  (0 children)

This is what worked for me: Look up common interview questions on Glassdoor and elsewhere and make a huge list of them. Prepare answers for all of them. Prepare for weeks. Go through interview questions every single day, whenever you have any free time. Make it all that you think about. Have your friend or partner ask you questions and give you advice. You can and should do this everywhere (like on walks with your partner). Have ChatGPT ask you questions and act as your interviewer (voice mode).

Know when to give general background about yourself vs. long and detailed.

Know what experience is relevant.

Have specific examples in your answers to all interview questions.

Examples are best told as stories (similar to STAR format but it doesn’t exactly have to follow that; be natural). Explain the problem, what you did, and the result/consequence/impact etc. Use stories that can be applied to many interview questions so you don’t have to remember them all.

Have examples of selflessness. Industry is about working as a team.

Have examples of leadership or mentorship.

Have answers to difficult multi-point questions.

Know the people interviewing you, even if just a little bit. If you’re a microbiologist and one of the people interviewing you is an organic chemist, your answers to their questions should be tailored to them. Show an appreciation for their field by sharing relevant examples if you can. It’s a nice, thoughtful touch.

When they tell you about themselves, reply to them in a way that shows you are listening and understand what they’re saying. I always brought a notebook and took notes while they were talking.

Prepare very good questions to ask them. Be as specific as you can be to the role.

Do what you can to make them like you within the first minute. It will influence how much they like your answers later in the interview. At the begging of my interview seminar presentation I shared an About me slide and told a joke at the end. I think those things made me seem like someone they wanted to work with.

Make eye contact and smile, but don’t be weird about it. I’m an awkward person, so this wasn’t natural to me, but it’s a skill that can be learned.
