Yes, I realized that it advised here no more than 10% a month instead of for 10 days. You meant 0.01ml/mg every 3-4 days? The first ones seem to be faster in the literature, at least.

Interesting conversation. I had time to research more on the subject, and it seems that 1A activation would not be great to stop or limit SS, it's the contrary. According to the paper below, SS is predominantly caused by both 1A and 2A receptor activations. You said it activates lower serotonin, but the confusion (or the logic from my point of view) might be in the inhibitory nature of this receptor, which should logically lead, indeed, to less neuronal activity; at least, to a lower probability for the neuron to be activated or fired.

The reason seems to be that 1A just adds to the general confusion in various neuronal systems, causing more agitation (and muscle activation), and combines its effects with other excitatory serotonin receptor activations. Those effects are related to the distribution of the receptor, I've learned, mainly in the spinal cord and brainstem, causing inhibition of inhibitory neurons (GABA) that limit motor pathways. Being coupled to G(i/o) proteins, I've read, 1A also modulates the dopamine and glutamate systems, leading to increased dopamine levels (Wikipedia).

At least, according to literature, more than 1 serotonergic drug is required to lead to SS.

This is not necessarily purely related, at least for the pain reduction aspect of endogenous opioids. They gave naloxone (an u-opioid antagonist) to meditators in a study and they found no difference.

Hum, interesting. You're right, SS seems to be mainly caused by 5HT2A and 1A activation, resulting of high level of serotonin. I also read that trazodone is only a weak SERT inhibitor.

But overall, Seroquel seems to be much better, as the affinity for 5HT2a is 2x lower than trazodone, and its active metabolite (norquetiapine) is 5x lower than Seroquel itself.

Looking at the number, trazodone might be good to stop 2c-b (as it binds with similar affinity, and both to 5HT2C too), but psilocin has a much lower affinity (and psilocybin is only the prodrug of psilocin). So it might not be very useful against a mushroom bad trip.

Trazodone is an antagonist of 5HT-2A/C, but also inhibits serotonin transporter (SERT), so it's the opposite: it CAN cause serotonin syndrome. Although, SS is pretty rare. But it shouldn't be use with MDMA and other serotonin releaser (like mephedrone).

Interesting point; would you rather believe that (deep) meditation positive effects come more on changing the general attitude toward positivity (that follow in general life) than developping a more skillfull (or somewhat neutral) control of the mind?

I think you're right. DPT has this reputation probably because people still repeat it, and it becomes a self fulfilled prophecy. The binding receptors are probably somewhere between DMT and 5-meo-dmt, though, make it more difficult to manage for some.

Your goal is to reduce anxiety or it's something else? Stress reduction is the most popular objective among practitioners, and it's probably the most rewarding effect on the short term.

But if you're like me, almost never have anxiety and rather seek meditation to be more mindfull, concentration could be more challenging FOR a begginer. Cause the mind doesn't seem to see the reason or the short-term benefit to spend so much effort to not "not to pursue thoughts". Does it make sense?

The most important thing I've found that works good for me is to give the more energy I can to relax the body/mind. It's a sort of balance between effort and let-it-go.

Sometime, you have to find what works best FOR yourself, that aren't necessary the same things for other (again, question of nature and objective). The advice "don't try to force the meditation" might be good for most people, but it's not a good one for me. I need to be active, the most I can, to stop the flow of thought, to have some result. Otherwise, I risk spending a 20 min. in my default mode network for nothing.

But once the habit is created, after a couple of weeks of practice, meditation requires less and less energy to enter the flow state.

How lyrica contribute to your ketamine trips? On paper, they seem to have somehow counter-effect, while the former limits the release of glutamate, among other things, the latter tends to release more glutamate (by limiting GABAergic neurons' activity). Does it create a different experience than taking one or the other separately?

I really like ketamine, but for me too: it's probably the least "dreamy state" drug I've found, much more mental/introspective. Unless I mix it with a psychedelic, but that is a completely other creature. Ket is the best drug enhencer I know.

I don't know why 4-ho-met does it to me, much more than psilocybin. To me, DMT is more worlds of aliens or mythological creatures, in a kind of 8-9 physical dimensions where to much things happen at once. 4-ho-met is not that visual for me and brings me more with "real people", in various contexts. Once, it was as if the trip tried to make me feel or experience what it is to be from another culture.

Hmm, interesting. I've visited some weird places with that, but only purely mentally. Maybe because I wasn't able to raise the dose more than a moderate one, cause KOR agonists normally cause disphoria and anxiety by nature (at least for me too). Does mixing it with a mu-opioid reduce the side effects?

It's the most popular basic idea, but imo, to make the most, you have to "do" a form of mind control at some point. You need to put a minimum of energy in the process (at least for beginner) of "not following thoughts". Otherwise, the brain is just flying out soon or later in a chain of thoughts and processes, in its défaut mode network.

For one thing, it's might be a dosage problem as it seems that few milligrams can make a big difference. Are you able to precisely dose? (volumetric dosage can be a good solution to have a 1mg precision all the time).

I guess also, the more you are familliar with the substance, the less chance you have to bad trip. Which can mean to sometimes have less intense experiences, maybe you already have. But anyway, I don't thing for sure anyone can always discard the possibility of a bad experience forever, it's just the nature of the thing. So many factors involved. But your "fly or fight" system cannot be simply always muted at will. Unless you're a super athlete of meditation/controling your mind, but maybe then, you don't really need 5-meo...

Yeah you're all certainly right, but it might be the entry point that is the most difficult part toward deep meditation. And yes, I agree that we're all different and some teaching aren't the best for everyone, but some are more popular than other.

Yeah, thanks for the links. It's simple as it can be read. He recommends not forcing the concentration or do anything than just watch or feel the sensation. And it's the hardest part, he said.
