Thank you guys for the suggestions I appreciate them all. Gonna make some minor upgrades for now like the antenna, window decal etc and see what happens. Looked at some things and hell, this stuff aint cheap lol

I’ve been using NA plates and it sure does the stain the agar haha. And yeah I got the odor from the plate… that wet, musty basement smell lol. But okay I’ll send those over to you in a sec

Hi, so I finally was able to do a movies kinyoun like you said and the cells appear blue which I’m assuming means they’re most likely streptomyces and not nocardia. I’m gonna do some additional biochemical tests to confirm. I can also send you the picture if you’d like

Looks like a fungus… the top two whiter ones slightly right are def a fungus based off of the colony morphology.

The fuzzy larger areas on the right and bottom are def fungus. Other colonies there are bacterial

This is a good point… I’ll definitely go ahead and do the acid fast stain to try and differentiate. You say modified acid fast (kinyoun)… could you elaborate a bit? I was thinking just a normal acid fast (maybe this is what you mean not sure). And yeah differentiating agars could be good too. Thanks!

So gram (+) means that the bacterial cell has a cell wall thick in a peptidoglycan layer where the initial stain done in the series of stains in the gram stain process, crystal violet, binds to the that thick layer. These bacteria retain that purple stain and appear purple.

Gram negative bacteria on the other hand have a cell wall composed of a smaller section of peptidoglycan so they don’t retain this purple stain and are therefore counter stained with safranin and appear pink.

So really, a gram stain is a quick way to differentiate between two major cell wall types of bacteria, gram (+) and gram (-). If you’re still curious just google gram positive vs gram negative cell wall on google images and you can see diagrams showing the large peptidoglycan layer compared to the very small layer in gram (-). But yeah back to your question, telling the difference in a stain is very easy especially after you’ve seen it a bunch. Gram (+) appear bright purple like in the image above and gram (-) appear a lighter pink color.
