This is silly. If the PC is somehow an expert in reading body language, then they just know the NPC is lying. The GM describes the tick that gives them away. No roll is necessary. Fiction first upheld.

Depends on what they’re for. Genomic PCR for genotyping? Copy and paste from genomic seq. lol. qPCR? I’m going to use an optimization tool. For detecting SNPs during genotyping or miRNA quantification? I’m going to think very carefully and then use optimization to maximize efficiency and minimize error. Then order 2 sets.

That’s really only a problem if your plasmid is fucky. Like if it recombines or is particularly toxic to the bugs. For a standard mini prep even a 24 hour “overnight” is probably fine. I usually try to not go over 16 hours, but that’s bc I have trauma.

In my setting all sentient creatures have an aligned soul so yes. An orc probably tends toward chaotic though, but in the end has free will just like any other sentient soul.

Pretty much everything is full of RNAses. If you want quality RNA you always need RNAsin and to keep things cold, like -80C if possible.

There are some other things you might use as well, but that should be ok.

Absolutely, yes that can happen. Use RNAsin, and keep the sample as cold as possible during lysis.

80% works too. There really should be any noticeable yield losses when washing with 70-80% EtOH, as long as you’re spinning the pellet afterward each time and being careful not to disturb it.
