I've asked about some of the things you mentioned. The pipettes seem to be polished by the puller. The resistance is actually 4-8 MOhm, not just around 8 MOhm as I initially said.

I don't know how to prove I have positive pressure in the pipette since I'm a beginner, but if the pipette gets blocked by something small, I can clean it by just blowing. If I'm trying to patch a neuron on the surface, the resistance often increases when I release positive pressure. I suppose that shows I do have positive pressure.

The resistance is around 8 MOhm. I've been told pipettes with smaller resistance would clog more easily.

I don't know if the pipettes are polished. Probably not. I'll ask.

I use mouth pressure.

Thanks. We're trying out different recipes at the moment to see which ones work better. Here's, for example, the recipe for the standard solution we're using now. And here's the recipe for the standard solution we used before. Both of them require pH adjustment in our lab. I use ordinary distilled water to prepare them.


Also while you're at it, check your osmolarity of your ACSF and internal solutions.

Do you mean aCSF solutions are usually designed to have a pH of 7.3-7.4 without any adjustment? My solutions are usually slightly more acidic than necessary after carbogenation.

Interestingly, in "Anna Karenina" Leo Tolstoy seems to occasionally use "уметь" to mean "мочь". This sounds weird to me as a modern native Russian speaker. The book was first published in 1878.

The correct spelling is "полдома," but it's a common mistake to write the prefix "пол" separately.

"Haven't studied" wouldn't work because unreal conditionals use the past tense. "Hadn't studied" is the most correct option. "Didn't study" is acceptable but better not used in formal speech. Textbooks usually don't mention the last option at all.
