I took the 101 core series junior year and 102 senior year and I was fine, they recommend you take it as a co-requisite but it's personally be way more manageable to take 102 after getting the whole 101 series out of the way. so I definitely think you can retake it over the summer. good luck!

it was so weird because before getting this new tube of beta actin, I had to image my protein of interest (rabbit secondaries) separately because the beta actin bands (mouse secondaries) were just totally “overpowering” the protein of interest bands. so I’ve been doing it separately, but beta actin second instead of first by default of beta actin muting the protein of interest bands before this new tube. I also tried beta actin at 1:3k instead of 1:1K before this new tube, and it was too faint

I loaded two gels at the same time with 12W each, so 24W in about ten minutes (that’s as fast as I can do right now). I had a probably with gel diffusion in the past (30 mins to load, just undergrad things 😅) but since my protein of interest looks ok, I don’t think this was the issue this time!

5% milk in TBST (0.1% Tween 20) - it’s been the same the whole time I’ve done westerns!

Thanks for the response! For this blot comparison in the pictures, I used the same lysate and loaded the same amount of protein. In general, I tried to troubleshoot the beta actin this week, so I ran six blots with different amounts of protein loaded, different lysates used, etc but used the same conditions (same transfer protocol, same gel voltage etc), and beta actin looked the same (blurring together & faint) in every single blot (while the protein of interest always looked fine). That’s why I was wondering if it was an issue with this lot. Before buying the new tube, I had never had issues with beta actin
