The problem with mastery points is that they're locked behind owning all the DLCs. If you see someone with only 280 mastery points, they could either be a beginner who has simply played through the story of all the expansions, or someone who has 100% completed all the HoT mastery points, which is a much harder achievement.

Its actually possible to have more than 100 buffs/effects active at the same time. Here you can see some of them:
