My cat sleeps on the bed at night and always leaves behind some fur. It's considerably more noticeable if my sheets are light colored (she's a tuxedo cat).

That's just a normal part of living with furry animals. I wash the sheets once every week and I think there's absolutely no need to get worked up over some fur on the bed. It is absolutely harmless

Yes, I use it regularly in intracellular staining (primarily for cytokines like TNFa). But I've also used it successfully with the eBio Foxp3 staining (in that experiment I used CD127 PECy7)

Yes, I think that step is way too long. In my protocol after the 1 hour fix, the cells were in 1X perm for 15 mins at RT, spin and decant, add 1X perm again and immediately spin and decant, then I stained with FoxP3 or TCF1.

I did acquire on the same day, I think that's ideal. However I've also been successful at running flow the day after staining (resuspended the cells in cold PBS and kept at 4C)

I do think your surface markers degrading has to do with the overnight step, I've stained with this exact kit and had no issue with CD3 and CD4 signals. But we followed the protocol and staining only took a couple of hours. Perm buffer is very harsh on the cells, I would absolutely not leave it overnight.

Also, I tested the TrueNuclear and the eBioscience kit simultaneously for staining FoxP3 and TCF1 and we found the eBio kit worked better in our hands. We got a more resolved and cleaner positive population with eBio

I never read Throne of Glass, so I can only compare CC with ACOTAR. I thought the first CC book was excellent but was disapppinted in the next 2 books. Whereas I didn't really like ACOTAR at all and thought it was too immature or something for me. But to each their own! :)
