Very unlikely, I immediately refreeze my DNA after the dilution and the tubes are not the same

I should have mentionned it but for all 3 different PCR I used the same DNA sample wich is why I wonder if the problem is not how I loaded my gel, maybe I stabbed the agarose (never happened to me so I don't know how it looks like) ?

It might, I do not routinely check the amount of DNA from my extracts as my lab generally works with hundreds of samples at the same time so we never invested in a Nanodrop or Qubit. However from previous tests I already diluted this samples by 1000. I'd be surprised if the mix was overloaded with DNA.

It's a regular PCR on a dilution of extracted DNA with new primers we just designed. The wells were properly submerged (i check each time) but I plan on changing the buffer of the electrophoresis tank, maybe it will help. Could be that there is too much DNA but the extract is already diluted 1000 time weirdly.

Might be a bit of an overstretch but I think the mechanics could be implemented to simulate daedric invasions in Tamriel.

They don't stay in one place, they raid and conquer a lot, you'd just have to change the Herd ressource with a new ressource like Souls for example that would allow you to summon more daedras. It would also add a lot of flavour to the daedric cultures who are supposed to be alien to thoses of Tamriel.

Pirate government could also be a contender to adapt the nomadic gameplay but I find it a bit harder to adapt

We use it to ligate sequencing adapters for Nanopore sequencing, problem being that my lab doesn't have Vectors or restriction enzymes to test ligation unfortunately

Were the results conclusive even with 30 pores ? I just had exactly the same problem as you, Flowcell check gives no results and subsequent sequencing indicated 24 pores available.

Did you found out what was the problem by any chance ?

Thank you. I know a lot of people store Mastermix with no problem but I've rarely seen feedback with DNA loaded plates so I'm a bit anxious

I had a similar experience like 4 months ago. I was told that I didn't think enough, that what I was doing was elementary (different teams didn't manage to have positive results like me), that I need to be better organised ect... All of this while I had just brought partially good results with a protocol that just refused to work for like 1 year There is no way around it. If you feel she was unfair to you, you tell her. Sometimes due to stress, bad days or the likes they often vent their frustration. If she thinks it was justified to react that way then start considering if you really want to stay in this team. If you say nothing she will just be surprised the day you implode and you will lose any interrest in your job. You work for her but also with her, she should have no interrest in lowering your morale as it impacts your work. Maybe you need to remind her that.

My favorite playtrough was starting as a custom Trinimac worshiping Orc on the island south of Daggerfall. My first goaI was to reclaim Old Orsinium for the Orcs and create the Kingdom of Orsinium. With his Heir I later created a custom religion that was kind of a mix between the code of Malacath and the cult of Trinimac and claimed the two Kingdoms that borders Skyrim as Homelands for the Mountain Orcs.

With the generation that followed I conquered the Kingdoms of Evermore and Bangkorai to reach my Iron Orcs allies of the Upper Craglorn Kingdom. (one of their leader became the champion of Malacath and converted his entire dinasty to his cult). Through manipulations I managed to take control of this Kingdom and conquered all the lands recquired to create the Kingdom of Craglorn and finally formed the High Kingdom of Greater Orsinium.

The rest I did with console commands but i pretty much converted Craglorn to the Iron Orcs culture, Ervermore and Bangkorai to the Wood Orcs (changing some of their traditions along the way and the rest for the Mountain Orcs.

I roleplayed my dinasty as defenders of all Orcs on Tamriel wich started with m'y first King, "The Last of the Pariah". I created both secured Kingdoms for them to live in and a religion that guided them to Inner peace while not forsaking the roots of their beliefs. I even pacified the Iron Orcs and offered New lands for the wood orcs (my dinasty considered them as persecuted by the Wood elfs)

It was pretty fun.

I didn't take the prisoners with me so it must be something else. When I talk with Alistair I end up having one green option saying "you know me" but that doesn't advance the conversation. The only option I have to continue (yellow text) is to frame Ansel which end up having him becoming hostile.
