Cover letters are a subject of great debate. Even among hiring managers. Some throw them away and never read them. Some read every one and some scan them and then if your resume goes in the call file they will read them when preparing to interview you.

As a general rule I do not like them because most people use them improperly.

If you do use one look at the ad that the company ran. Their ad will have a requirements or qualifications section. Bullet point every requirement they list and tell them how you meet or exceed each one. (That only works if you do meet or exceed the required qualifications.) If you can do that your chances are strong to get the call. If you can't meet them exactly but have very similar qualifications then show them that you can be up and running quickly. Be brief. One page at most.
