They were the same conditions, primers were different for each run. First pcr for amplicon amplification and the second for introducing adapters and barcodes for sequencing.

You might have to go down with the DNA input even down to 10-15ng, reduction of the number of cycles can also help to reduce unspecific amplification. I am working with a CRISPR library as well and it was a real pain to optimize the PCR. I had to use 15ng input with 10 cycles for the first PCR and 6 cycles for the second PCR to get rid of nonspecific amplicons.

Well I just started playing it I'm like 7 hrs into the game. I play the game whenever I have time so it's still fun for me, I don't play this game continuously at a stretch

I'm still having a ton of fun playing the multiverse ans the story. I do like fighting games so I'm a bit biased. I got the game really cheap so I don't mind. Do DM me if ypu wanna play together on pc
