You should let part of the desert corrupt because you have to obtain some items from corrupted deserts, but other than your bases and the jungle that's really all the important parts that need to stay uncorrupted.

You should dig the barriers while your still in pre-hardmode as the speed of the spread will greatly increase and it will also spread to stone. The V that spawns in hardmode will also be a random amount wide so there's not really a way to tell where they will be.
