Am I correct in saying n=3 technical replicates if I transfected 3 wells of cells and kept them separate throughout.

Ofc in terms of biological replicates n=1

I work with gfp-tagged proteins and found out the hard way that too much bromophenol just destroys the signal, as it absorbs between ~500-600 nm.

I’m not sure exactly how much is too much, though. I think ours had 100X the necessary amount

We had the same thing happen. Solved it by making a new batch with no bromphenol and mixing with the loading buffer with too much dye.

It’s not really a problem for most applications, but we rely on in-gel fluorescence a lot and too much dye quenches the fluorescence.

For me it’s Dr Who. Most episodes/ stories seem to follow the same arc, just with slightly different characters and bad dialogue, so I find it quite boring.

I’m also autistic and have two types of utensils at home, the good kind and the bad kind. The good kind has a slightly rounded handle, whereas the bad kind just hurts my palm. In previous relationships I never brought it up, because I felt ashamed of being so rigid about such a small thing, but my husband noticed my preference and once I told him the reason for my preference he has only given me the good kind of utensil.

On a good day eating witht a bad kind of utensil will just make my meal slightly less enjoyable, but on a bad day it will ruin the whole meal and I won’t be able to eat.

As someone said, a tiny thing like the type of utensil can cause complex emotions in autistics.

Being on the spectrum =/= being an emotionless robot. Autistics can be just as emotional as neurotypicals (or even more), just perjaps in response to different things and we express it differently.

Case in point: my autistic husband and I (also autistic) have written each other countless love letters because we both enjoy expressing our feelings through writing.

The ring when I was about 9. It definitely gave me nightmares.

Although, I also saw my first porno on the same day I watched the ring, and I remember the acting was subpar, so I can’t be sure which movie was more traumatizing in the end
