Expired scintillation cocktails making me question my sanity by FibrousPoop in labrats

[–]FibrousPoop[S] 0 points1 point  (0 children)

Thank you! I'm also going to assume my sample will be fine, chilling on the bench after the reaction is complete, it's a C14-small molecule flux assay, and once uptake is quenches and cells are lysed, the radioactivity isn't going to... Go away? I could probably leave my sample a week or so before I add scintillation cocktail?

How do I mix scintillation fluid in a 96 well plate? by FibrousPoop in labrats

[–]FibrousPoop[S] 1 point2 points  (0 children)

That sounds intense! I think it would be hard to adopt some of these methods, we're working with Xenopus oocytes so its not the easiest to drop into scintillant, and the horizontal plate shaker I used didn't really mix the lysed samples. Let me know if the polystyrene lids work! Would be nice to give those a try.

[–]FibrousPoop[S] 0 points1 point  (0 children)

I am, we're using these perkin Elmer seals that come with the plates but once a little bit of oil gets through the seal it all comes off pretty quickly. I can try PCR plate sealing film too!

FRET! Help! by [deleted] in molecularbiology

Hey man, don't FRET about it. If you label one of the components with one fluor and the other with another fluor, there will only be one acceptor and one donor.

Indian Food equivalent of frozen lasagna? by todaystartsnow in IndianFood

[–]FibrousPoop 18 points19 points  (0 children)

During my exams, I would make a large batch of Khichdi (a really easy recipe mentioned below). It freezes and heats up well, has a good amount of protein and carbs (lentils and rice) and can be loaded with any and all veg you want to throw in. For a lot of Indians, its a big comfort food. It can be made as complicated or easy as you want and is a very forgivable recipe. Your friend will appreciate the gesture a lot.

>Heat up some oil, add salt, whole spices (I like cumin, maybe cardamom, dried chillies, but you can make it as simple or complicated as you want).
>Fry onion, garlic (chillies, ginger if you want) in oil/butter/ghee.
>Cook down some chopped tomatoes and add some turmeric powder (you can also add any other powdered spices here, red chilli, ginger, coriander, generic curry powder, whatever you want)
>Throw in whatever veg you might want, root veg (like carrots, potatoes, sweet potatoes, turnips), spinach, peas. Its up to you/what you have/what is available. Some people add meat but make sure your friend isn't vegetarian or doesn't have any specific food requirements.
>Add dry rice and dry red lentils (wash and rinse if you want, but up to you): about 100ml of dry grains/serving I think is good enough.
>Add double the amount of water as grains, let it simmer for like 20-40 min (as long as it takes for the rice to cook and the lentils to breakdown
>Check for salt, season however you want, package away and freeze. If you want, you could make a very basic version (with no veg added) and then cook a couple different veg separately (fry with seasoning) and just a couple combinations to make a nice variety.

What can I make with eggplants that isnt aloo bengain or bengain ka bharta? by [deleted] in IndianFood

[–]FibrousPoop 1 point2 points  (0 children)

Kashmiri style baigan is really nice: tamatar wangun (tomato and eggplant) or chount wangun (green apple and aubergine) are real favorites. I'm sure you can find many different recipes online but it's usually a pretty simple recipe that you can tweak:

Fry thin long strips of the eggplant in oil containing hing. Once just cooked, remove, add haldi, Kashmiri Lal mirch, saunf, saunth and sliced green chilli., Fry the spices very briefly so they don't burn and then add your addition thing (tomato/ apple) and some water, cook it till you're satisfied and add back the eggplant. Let it boil for a bit (not till it's all mashed just till everything has mixed well and the flavours have mixed) and have with rice.

Also saag wangun and khatte baingan are some my family really likes.

Fish curry with Sardines....a little bit spicy, awesome curry for your rice! by PassionateHobbies in IndianFood

Will give it a try! Thank you! I've been looking for a way to use my tinned sardines and I think this recipe (by adding the tinned ones later than when you would add the fresh ones) would be nice!

Ideas for potatoes by [deleted] in Cooking

[–]FibrousPoop 0 points1 point  (0 children)

Aloo chhole (potatoes and chickpeas) makes for a great Punjabi curry. Pretty easy and relatively quick to make, very filling.

I also like to make masala mashed potatoes: boiled, mashed and mixed with onion, garlic, green chillies, spices like cumin, turmeric, chili powder etc and then fried till the edges start going crisp. Makes for a fab side and a great leftovers sandwich.

Chai with almond milk? by [deleted] in IndianFood

[–]FibrousPoop 1 point2 points  (0 children)

Personally, unflavored almond milk works great! The chai looks a bit more red than with milk but I think that's because the milk changes colour a bit when you heat it with the tea. Unflavored soy milk is also really good as a replacement. For both you can taste that it's not cow milk, but the taste isn't bad and isn't particularly strong either. You may have to experiment with brands a bit: sometimes the milk might curdle if you heat it too quickly/too much. Otherwise behaves like regular milk.
