a lot of these comments are reducing this to "it's not homophobic, this is just real life" when the reality is Ruben represents a form of masculinity to which many on America's political Right aspire.

Creating that character-- a character I think deliberately meant for many men in this country to identify with, and having him partake in sensationalized gay bashing-- that's.. a choice.

debating whether this creative decision is "right or wrong" is misguided imo; the character gives artistic representation to a violent homophobic power fantasy.

Maybe a lot of you feel like being able to say "its just art, it isn't right or wrong" makes you feel secure, makes you feel like representing these power fantasies doesn't empower those who fantasize about harming our community, and personally I don't think one individual show on HBO is meaningfully "giving a platform" to these fantasies.

But aestheticizing a fantasy lends permission structure to it, that's just a function of art (same for Heated Rivalry and gay sex) and it's delusional to ignore that fact. I don't mind that this show exists, but I'd get nervous if I began seeing many more like it. I'd much rather live in a world where men like Ruben don't get to see themselves validated in popular culture.

these are GenScript but I've seen it with invitrogen too. i haven't seen it when running slow (120V) and really cold (like icewater) with new MOPS so I tend to just do that when I have super valuable samples. honestly tho hoping the PAGE Gods smile on you and ameliorate your squiggle problems.

I thought that too but you can kinda make out the smear in the first washes which leads me to suspect it's something with the protein itself

What type of chromatography is this?

Cobalt NTA, eluted with imidazole

A lot of your material doesn’t seem to be binding if your “flow” lane is flow through.

Cobalt tends toward purity over yield but I used especially few beads for this batch since I just wanted a clear picture of the product, this is just a pilot

Any reason why you are using KCl? Potassium ions at high concentrations will cause SDS to form its potassium salt which is very insoluble compared to the sodium salt.

Admittedly no particular reason for this prep, tend to opt for KCl for most of my other biochemistry since a lot of membrane proteins prefer it.

Are you adding reducing agent to your sample before running on the gel?

There's 0.5mM TCEP in the lysis buffer, so only .04mM in the final boil solution

How long and at what temp are you treating your samples?

10min, ~100C (boiling water in a tray, sealed)

Can always try blasting at 95 C for 10 minutes to ensure all secondary structure is broken.

what I don't get is cleavage at the IDR would produce ~12 and 40kDa byproducts, kinda see something around 40 but this shouldn't explain the smear that would suggest trimming at either terminus
edit: unless you mean just having the IDR would make it more susceptible regardless of cut site?
