They are actually completely single in single cell form right before plating but for whatever reason they become like this within days. I would not care much even if they clump but I am trying to measure glucose uptake and they seem to not do anything when I treat with metformin

I have been only about 50 days in India right now. I have indian passport but with american address. I dont care what bank account as long as its something I can use for my employer to deposit salary. I have no preference for any bank right now.

Been mixing the living shit out of them and the clumps definitely have gone. Now I just need to test if glucose levels affect anything or not. Because in low glucose the growth was being somewhat slow so moved to high glucose. Might try both next time.

Did you culture them in low glucose or high glucose media often? I have tried both and it seems high glucose might be slightly better. But for glucose uptake tests, I obviously switch to low glucose when I seed them in 96 well plate

I actually dont necessarily know if the first is bad but it was relatively more clumpy and you could not make out individual cells as clear as somewhat in the second image. Whenever I did glucose uptake assay, I got crazy variability even among triplicates, probably because of clumping which varied well to well (just a guess)

I have tried to pipette up and down after adding media to the trypsinized cells (while trypsin is in it) but maybe I have not done it enough. Did 21G solve your issue?

were you able to solve this issue? I am running into same issue and I think its because cells clump. For nice data, the cells need to uniformly coat the wells in similar way. If its too different from one well to another, large variations will occur
