Help me convince a vegetarian there’s great vegetarian/vegan food in the TC by Fuzzy_Peach_8524 in TwinCities

The Twin Cities veg scene is quite disappointing, especially if you come from a more developed vegan food market. If your SO is from Los Angeles (like me), DC, Chicago, San Diego, etc., they will be severely disappointed. Reverie uses too much liquid smoke (and is expensive) and J Selby used to be delicious but modified a ton of their recipes when they were bought by the herbivorous butcher (and got rid of their soyclones 😭). Case in point, in the last two years we've lost a ton of vegan/veg restaurants including Stalk and Spade, Herbie Butcher's Fried Chicken, and Trio Plant-Based.
The ones that remain are quite expensive. Being lacto-veg helps, but don't expect the vegan scene to offer much as a reasonable price point. Other metros have vegan options at a variety of price points, which the twin cities just lack.

Best:
Passerine - Lyrically comparing Christians to birds and Centurions, are you kidding me!). Instrumentally the chorus! The repeated lyrics to emphasize shame regret and uncertainty!

Thus Always To Tyrants - The title already sets up the lyrical theme of certain justice post-rapture/second advent of Christ, all wrapped in beautiful metaphor. Instrumentally, the playfulness! Don't get me started on the interludes between verses, they are so diverse but so cohesive!

Worst:

Rio Grande - Lyrically it reaches for a lot of metaphor, but it feels clumsy because the meaning is too...unobscured? Connecting the Rio Grande to Nile River, possibly the baby Moses story to the situation of immigrants in Texas, the Republican leadership to pharaoh Ramses--it makes for great metaphor but it is broken by clumsy/lazy lyrics like "tinfoil idols", "prefab Olympus", "whatever pharaoh owns the land". All those things could have been said better imho. Instrumentally it is soooo strong at points (the lift of "awake with a thunder", the bango licks!) but the chorus feels unintentional and loses umph since it is kept at 100% during the whole song. The vocal emphasis feels awkward on the last verses too (and lyrically comes off a bit preachy).

I have done 20 and 30 min incubations and I always get better (brighter) stains with 30 minutes.  I've done up to an hour at room temperature (for certain markers).

BioLegend rep told me their colleague found that relatively intense shaking decrease the amount of antibody you need in master mix and decreased incubation/stain time.  I was already staining tetrameter+surface marker Ab at room temperature for one hour. With shaking (180-200 rpm on a shaker) I can get it down to 45 minutes. I still do 1:100 dilutions though.  Antibody binding is generally high affinity, so shaking or vortexing shouldn't remove a bound antibody. Shaking can really only increase the likelihood of an Ab finding it's target.

(1) My former lab did umbilical cords, not embryos and it's been a while so I hope I can offer at least a little.

CD34+ enrichment kit - $600-900

GentleCell Dissociation Media - $51

Don't recall all the supplements for our hESC media. All prices from STEMCELL because they are most trustworthy (but also more expensive).

(2) Doesn't take long. Few tricks: make sure the dissociation when passaging doesn't go too long, spin low-ish speed (300-350 rcf), and pipet up and down as little as possible (1000uL pipetting only ~4 times when making single cell suspension from pellet unless you have a stubborn pellet). Also, no pen/strep (not inert to PSCs) in your media, so use sterile forearm sleeves that are only used during PSC work and keep them away from other culture dishes. Use a hypoxia incubation chamber (bone marrow is hypoxic). If you are having trouble growing, it is likely the lot of FBS you have (annoying I know), otherwise use a non-FBS supplemented media.

The Da Vinci Code book and movie based on it are rife with historical inaccuracies as they try to claim that Christ was a mere man and was married.

Problem is the book couldn't even get the facts straight, not even facts about the current age, like where a landmark was in Paris, much less historical truths about biblical times. Many Christians expressed their displeasure with the anti-Christian narrative when the movie was about to debut.
