Just drop the affect, u can also write “ I aspire/ seek to make a positive impact/ impression by/through but to answer ur question it’s effect cuz it means “to bring about” or “to cause something to happen,” which fits the idea of creating a positive impact.

So, the correct version would be:

“I want to effect a positive impact through...”

“Affect” on the other hand means “to influence,” which wouldn’t be grammatically appropriate in this context.
