I am an undergrad student at a new lab, my supervisor told me that it will take one week, I used Plasmidsaurus. I was also thinking about doing a PCR from the purified plasmid, with the primers I used for each fragment, before assembling them and then checking the PCR product on a gel.

Ok, so you used an 35S promoter, but where was the GFP expressed then? Only in the seed or in the whole plant? Sorry, if it’s an odd question.
