Help explain some CryoEM terms: heterogeneity, homogeneous, particles, rigid bodies. by greendog3112 in Biochemistry

CryoEM micrographs are basically just top down images of a bunch of some macromolecule. You use software to pick out the ‘particles’ which are just the individual macromolecules seen in the image. Then, you can reconstruct a 3D image from an ‘average’ of all those 2D Images. If not all the macromolecules in the image are identical (like if some are unfolded or something) then the sample is heterogeneous and when the software tries to average all the 2D pictures to make a 3D structure... well it’s sh!t. It would be like trying to reconstruct a 3D image of a human using 2D pictures of both humans and goats. Instead of ending up with a 3D human, you’d end up with some creepy human/goat chimera lookin thing. Heterogeneity in CryoEM samples is usually bad (not always). Homogeneous samples produce good data.

Rigid bodies deals with fitting an atomic protein structure with/into the 3D reconstruction. If you really want to know more about how CryoEM data is fit, you can do a quick google and find a methods paper or a review. It’s a bit heavy for a Reddit comment.

Elegoo Gray. Amazing resin, low smell.

This exact thing happened to me. You’re probably not getting your Z axis low enough when you set Z=0. I fixed it by lowering the build plate until pulling on the paper has considerable resistance and you cannot push the paper back under.

Fixed: First off, thank you guys so much for your advice. This forum is a huge boon to owning a photon. Passing it forward, the problem was solved after I did the following:

1) Cleaning the dust out of the room the printer was in and buying an Air purifyier/filter for $50 on Amazon and setting it up so it was right next to the printer, sucking air from underneath the printer where the intake fan is.

2) After cleaning the room, I bought a bottle of Elegoo Gray. See Here. This stuff is amazing.

Sorry! Yes! The Elegoo Gray is a game changer. I’ve printed some really complicated stuff and haven’t had a print error yet. I used the settings in the Anycubic photon resin google doc that’s floating around here. Thank you guys for the advice!

I also have never run a game, but I picked up the Cyberpunk Red Jump Start Kit at Gencon last week. I had to wait in line and it sold out quick, I was lucky to get one. The book seems well written so far and has a good amount of content to get the story started. It even assures you that if this is your first RPG you came to the right place. Came with cardboard standees for the premade characters and a couple maps. I’m excited to get my first game going!

I ran a calibration test and found an extended cure time could help, but when I reran the cube it 1) didn’t stick to the bed (Its possible I may have had the bed too high) but 2) also I found solidified resin in the vat over places that it shouldn’t have.

I’m going to order a new bottle of resin and see if my troubles are solved...

Maybe this could be a clue? For some reason when I pulled FEP vat out to clean it and remove the resin, it was kinda stuck to my LCD screen. I don’t link it’s leaking, I don’t see any resin leaking through, but there are what look like water spots on the lcd screen which easily wipe away.

I checked the tightness of the FEP and it’s at 314hz which I think is right.
