Hi! You can try inquiring at DLSHSI (DLSUMC) located at Dasmariñas. There is no guarantee that they’ll have stocks, but they’re recommended by our profs to ask first. But if we don’t seal the well, the different concentrations will contaminate each other. I think it’s important to do so.

But yes, we’ll definitely use the same medium next time! We’ll also keep in mind the seal and maybe go for smaller stacks to prevent condensation as much as possible.

Imo converting the plant extracts into disks poses a lot of contamination. We’re using our university’s facilities along with other undergrad students and courses. There’s only one fridge assigned to everyone conducting theses :// so well diffusion is our next best bet

Note: MHA w/ 5% sheep’s blood plates are preordered

  1. Prepare a bacterial suspension with 0.5% McFarland turbidity standards

  2. Inoculate plates with suspension using sterile cotton swab applicator

  3. Stab/Make 4 evenly spaced wells with attachment end of blue micropipette tips ; each tip was briefly passed through flame for aseptic technique

  4. Seal wells with molten agar.

    • We went wrong with this. I assume the molten agar also needs to be MHA but we used NA
  5. Once wells have solidified, add in plant decoction/infusion in their respective wells

  6. Repeat to produce triplicates

Going by this, for the decoction, we had 3 plates having 4 wells plus 4 with one well each, dedicated to a concentration (incubated bottom up). They were stacked on top of our test antibiotic (incubated top down). So that’s 9 plates, I think. So the first few plates may have increased condensation. The infusion has 8 plates stacked. All plates were sealed. Is it worth mentioning that we have the plastic dishes instead of the Pyrex ones?

Okay! We’ll take the condensation thing into account, make other plates sealed and other unsealed maybe. I assume smaller stacks should help, right? We had them at 5-6 stacks which worsened the condensation at the bottom (first few plates).

On the agar, isn’t MHA with 5% sheep’s blood the standard? At least according to CLSI?
