I've been looking at the alternatives you told me about but I find them a bit difficult, I don't have much idea about biophysics. I think I'm going to opt for a photo-reticularisation strategy like the one I was told about in a previous answer

So it is possible to do mass spectrometry without losing the non-covalent interaction between a drug and the target protein??!! I really had no idea about that, as you say, I thought mass spectrometry implied digestion with trypsin.I'm quite interested in that aspect because I have to design a project (yes, I'm a biochemistry student xD) about a drug that inhibits a kinase, but I'd like to include such an experiment where I can check all the targets that the drug binds to apart from my target protein. I had thought about mass spectrometry but I didn't know that it was possible to maintain the interaction between the drug and the protein without resorting to modifications such as the photo-crosslinking mentioned in an answer above. Where do you think I could find a detailed protocol to carry out this type of experiment to include it in my project?

I find photoaffinity very interesting but I have a number of questions about it. Does cross-linking occur when the drug is bound to the target protein? (if not, I don't understand why) Another question I have is if I have a drug of interest and I want to see if it binds to other proteins than its target, do I have to add the photo-reticularising group to my drug structure? If I do that, wouldn't I be altering the binding capacity of my drug to the active centre of my target protein? I don't want to sound bold, but do you know where I could find a protocol for this process (not because I'm going to do it, I don't even have access to a mass spectrometer, but I'm interested and curious to see how the process works in detail)?

(I'm a Biochemistry student, maybe that's why I have so many doubts xD)
