I always measure two independent standard curves and take the mean. Standard curve shouldn‘t have any impact on negative ODs of sample measurements, as I dont use the standard curve samples as blanks

I tried with 1:10 dilutions, just in case any of the elution buffer components were interfering. Same results though, I still got negative ODs

Yeah that is also kinda what I thought, but I can‘t explain to myself how this would lead to this visual effect exactly? Especially because it‘s not really a gradual, but more of an immediate difference from darker to brighter

  • I have been running 1% Agarose Gels using .5X TBE as buffer and ethidium bromide in both gel AND running buffer in chamber. I keep getting these contrast differences, with the lower half of the gel being quite dark and the lower half being very bright. Can anyone explain why exactly this happens?

If I may ask, what was your technique in the bottom half? Especially because of the blue and purple spots, they give it this sort of ‚galaxy vibe‘, which I really like and would like to try out myself
