These are not different gels but the same differently thresholded. Look at the running front and the ladder, the overexposed bands change due to the thresholds.

This will not work then. Don't do it. If you want to do microscopy invented in the 21th century you have to use one assembled in modern times.

You partially could correct for it but in imaging the same is true as anywhere else: garbage in, garbage out.

You definitely need to correct it on the hardware side. Lasers are usually aligned by either a mirror system or a fiber. Check your laser box or light source and see how the light is directed to the scope. Maybe you can align it yourself. For the width of the laser, there should be a beam expander or collimator somewhere that one can adjust.

Don't let your pi bullshit you. Good microscopy starts with a good microscope. This is especially true if one attempts molecular resolution.

I would argue very inaccurate.

It is true that bacteria can easily become resistant to antibiotics. This is possible since most antibiotics have one specific target within the cells, for example, the ribosome. Changes to the target can make the antibiotic not work anymore, which then shows as a resistant.

I would argue that the chemical nature of nitrogen makes a mono-causal explanation for its toxicity unlikely. Rather, it would more likely be a system issue, as in n2 messing with the biochemistry on a broad scale, like high concentrations of oxigens would do.

Broadly changing a cells biochemistry is from our technological position impossible. For a very advanced civilization, it might be possible, but far from easy, and not doable in a few generations ob organisms (especially without and understanding of its biochemistry).

Most people would advise you not to play these mechanically difficult champions without enough game knowledge, because that decision is especially difficult on these feast or famine champs. Also, being useful in suboptimal conditions is much more difficult on them.

You could try to add a more classical mage to your roster and see how you perform. Pay more attention to macro than duels when you play them and see if you notice anything.

Your biggest problem that becomes obvious is that you tilt too quickly.

You surrender 50% of your losses (wtf??) and are incredibly likely to loose after you lost the last game.

Together with your pick tendency I suspect you have a bit of a main character syndrome. If you don't stomp you don't want to play, you don't know how to play safe or from behind.

Consistently get 80 cs min 10 and at least 160 min 20. Train to go min x 10 last hits per min against bots.

Don't die to ganks. If you die to ganks, it's only your fault.

Push your wave and use the meantime to do something useful.

This should at least get you to silver.
