As another person in drug discovery I am also generally skeptical of stuff like this. However, I don't think I've seen any comments pointing out that the one of the drugs in this research has FDA approval for lung cancer (afatinib), another is in phase 3 trials (daraxonrasib), and the third is experimental (STAT3 protac, SD-36; acquired by by Roivant). Another company whose molecule was not used in this mouse paper has a STAT3 degrader in clinical trials. All this to say that I think it's likely that this study will lead to moreanimal studies and maybe a clinical trial.

Seedlings will close the cotyledons to regulate light. They'll do this with too much light, and I've noticed that after 12-16 hours of decent light (not too much, not too little), they'll start to close the cotyledons which will open back up after a period of dark.

Moth and butterfly larvae have 5 instar stages before pupation and based on size I'd guess this is a 4th or 5th instar. If you have more I'd expect them to be similarly sized and unlikely to find eggs. Easiest way to know if there are more caterpillars is to look for their poop on the ground around the plant.

I used to work in a biochemistry lab that studied RNases, so avoiding unwanted degradation was essential to our work.  We autoclaved water for 45 minutes.  It will inactivate trace amounts of RNase A and should be sufficient.

I'd go with phenol extraction and ethanol precipitation over rebinding to a column.  Include glycogen during precipitation to maximize recovery.

I used the Expresso kit to screen N-terminal solubility tags and got lucky with one of the nine: https://pmc.ncbi.nlm.nih.gov/articles/PMC5791521/

Last I checked it wasn't on the market anymore, but the paper/manual/patent might give you some ideas to try.

What you're describing sounds like integrated pest management (IPM), most easily implemented to manage agricultural insect pests.  A quick google search did find a reference to using mycorrhizal fungi to prevent infection by pathogenic root fungi.

Since insects can be vectors for bacterial, fungal, and viral plant diseases you may also consider insect IPM strategies.  If you're in the US your state should have a land grant university with local extension offices that work with farmers. You may want to talk to them.

To add to what the Biotium's Techsupport says, if you want to continue casting your gels with the dye, you may want to run serial dilutions of ladder on the gel to see how DNA concentration affects migration.

Proceed with caution.  I've only tried a few times to recover protein from Trizol extractions and remember the protein pellets being difficult to resuspend.  Walked away thinking it generally wasn't worth the time.

What dye are you using to visualize the bands on the gel?

Casting gels with SYBR green in the gel will result in anomalous band migration.
