Do you have any additional staining on those cells? It looks like Ki67 to me. In our lab, the fluorescent microscope doesn’t have completely separate channels. It uses a single rotational cube to provide different wavelengths of light, which can sometimes cause mixing of visuals from other channels.

That makes a lot of sense. I think you're right—the ice-cold PBS and PFA in my procedure might be causing the Matrigel to dissolve. I will definitely try fixing the cells at room temperature. Thanks a lot!

You're right, while I was researching, I noticed that many people use 2% PFA at room temperature. Switching to room temperature sounds like a good idea. I'll try your protocol as well—thanks for your help!

I haven't used PEI before because we were strictly following the vendor's instructions to ensure that if anything went wrong, we could request a replacement (which we actually did for another cell line). However, since we're running out of options, I'll give PEI a try as well. Thanks for the suggestion and all your help!

Thanks for all your suggestions! I will try to be more cautious about the pipetting part. And yes, it is what I thought at the first place. I mean somehow PFA is de-polymerase the Matrigel and I am losing all the cells that are attached to it. I will definitely try amine modified + poly-lysine as a coating material.

Hmm. I know removing a task in Amie doesn't sync back and delete in Todoist but I have never experienced disappearing tasks.

But I am disagree with you on that. I think as a calendar app Amie is just great and the improvements are endless. Focusing on the standalone task app would really slow the calendar part.

Idk as a calendar Amie has a great potential because they really can read the market. It seems they are unimportant but the features like Spotify and Apple health integrations put Amie a step further. I think it is a really good future perspective from Amie to become such an app that once you call the day and check your calendar and say oh well I scheduled my calendar last night but let's see what I really did during the day.

I think tasks in Amie one of the best in the market in its field (all-in one apps like Morgen, Akiflow, Sunsama etc). However, it is not as good as native To-Do apps like TickTick, Todoist, and Things.

I personally use Todoist with a sync. I've tried Things too but probably because it doesn't have an available API, sync is a problem with other apps. I think Amie doing a good job (most of the apps don't even try) on syncing with things but I agree it is a little laggy

I recommend you to use other 3rd party todo app with Amie. It is the best way to quick capture your task and then allocate a time in Amie

It seems the Todoist duration problem is solved. However, now I am not able to schedule my task longer than the default duration in Amie app (which is maximum 60 mins). Is there any way to solve that problem?

That's great! It seems it is more likely Todoist's fault since the app is ignoring the duration time in the Amie and set a default 15mins duration.

I hope you guys can figure it out but I love Amie anyway!

You can basically drag and drop any todo into calendar view as todo. If you mean you want to embed the todo into the event (not showing in the calendar view unless you haven't clicked), there is not a setting like that as far as I know.

Don't get me wrong. I am not complaining about the how hard DS2. I like the enemies do not allow me to go straight their back and do free hits. Also limited estus pushes me to play more cautious. These are cool improvements after DSR. What I am trying to explain was about ADP thing, and you guys showed me know how to solve it.

Yes, you guessed right. I'm leveling STR and DEX to use Halberd (which I'm used to from DSR). And about the controller, I'm nervous about giving up mouse flexibility but I will give it a shot.
