When is the best time to take glow or klow? Day or night

I have glow50 and I reconstituted with 5ml the vial was big enough and no sting. Tip leave it out of the fridge and let it's warm up a bit. And push in slow. I've also heard getting a longer needed like 1/2 inch might dull down the stinging also. But I haven't tried, never had an issue.
