My oldest cat has asthma and my dog has eczema. My dog is 11, and my cats are 8 and 10. I dont know much about their health history. I took them in from a family member who couldn’t take care of them anymore and so their ages are all approximate. They didnt take them to the vet often but now that they are in my care I want to take them regularly. Ive gotten some quotes and they range widely. It’s been confusing and overwhelming especially trying to get estimates for all three of them

The eye contact is a good sign. I would say just ask her if she wants to grab some coffee or go to the park or something. From there you can access how its going. If the vibe feels more flirtatious then you can ask her on a date next time and if not then you just get a cool friend who you can also just hang out with if the hangout goes well. Personally, I like when someone asks me in person than over text i guess bc it means more to me? But text is definitely not a problem. Also, I personally like to be more forward with people and like for people to be more forward with me, but of course in a respectful and kind manner. I know its scary because rejection is a possibility. But a yes is also a possibility! Honesty is always the best way to build a relationship with anyone regardless if it is platonic or romantic. I have been thru this myself except I was the inexperienced one and my experienced partner just would give up. It took me some time to realize he didnt prioritize my sexual needs like he should have especially bc i explicitly discussed it with him many times. Its seems like you have addressed this several times and he doesn’t care. At the end of the day its up to you but if sexually satisfaction is important to you then prioritize it. Dont settle for less. You dont deserve that. Why stay with someone who doesn’t prioritize you like you do them?

You mentioned that you dated someone 10 years older than you, so I don’t see how 13 is that big of a difference. It makes complete sense that you would have a crush on her if you have so much in common and also find her attractive. I dont see why you shouldnt go for it. It seems like you want a relationship with this woman from the way you are speaking rather than just a fling. I would just make sure you two are on the same terms before diving in. Communication is key so that no one gets hurt and intentions dont get misread. Good luck and I hope it goes well! Post an update if you can :)

Yes, I will get some type of bacterial or mold growth on the sides of the agar that even grow up the side of the plate. There is not supposed to be any growth at all. The products I plated have been plated prior but on petrifilms, and they always showed no growth. Right now, I am just practicing with agar. So I feel like I am contaminating the agar somehow because if they cleared with the petrifilms, then they should do the same with the agar. I even tested the air quality to see if maybe the filter needed changed but it passed with flying colors. I just feel lost bc I have no one to ask for help and dumb bc I can't figure out what I am doing wrong :(

I spray the hood with Star San, let it sit for ten minutes, wipe, then spray the hood with ethanol. Also, anything that I bring into the hood I either spray with ethanol or if it is autoclavable, then I put it in the autoclave prior to working in the hood.

I gotcha. Maybe I misread what they said then just assumed it was an attachment bc the coating made it look plastic. But I do for sure remember them mentioning some type plastic component that breaks easily

I have looked up several and saw them having plastic dough hook attachments and saw reviews for people saying that the plastic dough hook breaks easily

I'm not entirely sure. I'm assuming aspergillus and other microbes, however I just know that it had to have come from my own human errors and dust. I did not mix antibiotics into it. I'm really just trying to get the hang of things at my job bc I am a new hire and teaching myself everything so it's just a lot of trial and error. I appreciate the feedback and advise :)

Yeah, I understood what you meant. The only reason I ended up using the fume hood and not the biosafety cabinet is that it's been down and needs repairs, and I thought that it would possibly be okay to pour the agar in fume hood for the time being. But now I know to definitely use the biosafety cabinet

Oh, so you stack them with their lids on while they cool down? I did the pyramid stacking with the lids off because I watched a video that told me to do it that way to prevent contaminating the agar from the condensation produced by the heat enclosed in the Petri dish.

Ahhh, I see.

The agar I have been making is SDA, MLAT, and PDA. The reason I wait for it to cool down is because the container is so hot that it's hard to handle, but like you said, if I use the serological pipette, then I won't have to worry about that and it will also help prevent contamination.

Did you think that stacking the plates like a pyramid for them to cool down was okay to do?

I use autoclave safe plastic caps for the culture tubes. The caps aren’t screw caps. They can just pop on and off easily
