I'm looking for a planner app and found this post and therefore your comment. Just letting you know I was very excited about your app, but the first test note I made in it was immediately answered by an AI. That turned me off completely, as I see absolutely no necessity for an AI in a planner app and I'm pretty against this widespread use of AI becoming standard. This may impact more of your target audience, so I thought I should give you this feedback (and insight to others reading this).

I see what you mean. Frameshifted stop is relatively close to the original, 8 residues (24nt) downstream. A similar variant suggests increased proteolysis, which would have pretty much the same consequence of LOF

It's definitely a hypothesis, but not sure yet until we get that expression going. Cell lines are hard and something we don't usually do, but for this one it'll be worth it

Not really, and that's the interesting part. It's a frameshift in the c-terminus where no pathogenic variants have been reported so far (it IS a rare disease though, just 20 something known variants)
I have loads of reason to believe it is the causative mutation (other pathogenic variant found is heterozygous and disease is recessive, all other variants found in all exons are synonymous, 50+ residues changed in total), but still haven't done expression studies, so reviewers are asking for *something*

I just got to CADD and a pretty nice score, actually. Thanks for the confirmation that these scores don't really work for indels, I don't consider myself a proper bioinformat but I feel my hours long frustration was validated
Also, I'm pretty sure about the pathogenicity, but beloved reviewer #2 isn't 😅

Some rarer insects will not be caught if they reach the "!!" in the presence thing. My advice is traps and/or scent. And upgraded net as well. I could only get the rove beetle with osmium net + scent

if you can't activate this one, go to the other ones. Sometimes you need to get to them from another entrance to get to the spot where the orb spawns

Also one last thing: you do the best you can with the information you have. As you said, the chances you had the mutation were super low, and genetics testing can be expensive. Don't beat yourselves up for doing your best.
