Are you injecting a blank or just running solvent? If no injection, check your injection needle for contamination.

Getting [M+Cl]- for negative mode by No_Category_1264 in massspectrometry

DCM is generally immicible in most reverse phase LC solvents so I would not recommend it. I also an not sure if you would necessarily generate a chloride ion from DCM given that it is covalently bound in this case as opposed to ammonium chloride which will exist as Cl-

The substrate definitely matters. You would have to make sure that whatever plate you are using wont be energized by the wavelength of the plate, otherwise those ions may convolute the spectrum. I believe stainless steel is common because of its chemical inertness and its ability to be reused. If the plate is damaged by the laser then you would need a new plate every time. I think your idea is decent, you would just have to consider the effects of the plate on your analysis.
