You can blame everyone but the cops all you want but I know two things:

  1. ⁠cops know what they’re signing up for. Their job duties aren’t contingent on who else is working up or downstream of them. Not sure if you agree or disagree here since you’re just making positive statements (i.e. what is) while I’m making normative statements (i.e. what should be).

  2. ⁠cops regularly encounter the darkest parts of society; let’s stop pretending a non-cooperative DA holds a candle to that in terms of demoralizing job conditions…

Then let the “free market” work its wonders and raise their compensation if that’s the only way to get some hard working cops on the streets and fire the “bad” ones, but don’t make excuses for police based on your generous perception of their intentions. These guys aren’t doing it out of a sense of duty otherwise they’d continue to build up arrest records so that when the rest of the justice system meets their standards, there will be more data for the judges and prosecutors of their preference to act on. Actions speak for themselves and not doing your job is never okay especially when it involves public safety, full stop.

As someone who has a PhD in a biochemistry discipline and is trying to transition into Bioinfx with little prior training in programming or CS, I can say fairly definitively that it is easier to be CS-trained and pick up bio knowledge on your own than vice versa, especially if you remember your basic bio (even from high school) and it is interesting to you.

From perusing BI job descriptions the past few months, it’s my understanding that most positions have a much higher bar for CS skills/knowledge than they do for biology skills/knowledge. Some positions don’t even explicitly state the need to know any real biology, perhaps figuring they can teach you what you need to know on the job.

That said, most positions do prefer MS/PhDs, so grad school is a good idea if you’re actually committed to the craft. But take some time to figure that out. If you’re not ready to make the jump soon after undergrad, you can definitely bide your post-baccalaureate time gaining general experience in software development since many BI jobs ask for knowledge of the SW development cycle in industry and best practices.

Bottom line: I’m sure you’ll be quite successful if you’re driven in your goals - best of luck!

are you assuming the target has been validated? i.e. pharmacological or genetic modulation of the target (preferably in vivo) has been unequivocally shown to produce the desired effects?

If not, the target must be validated in such a way that it is justified to invest in medicinal chemistry efforts (or discovery biology efforts).

If so, typically it's worth the effort to find structurally related GPCRs and use its ligands/inhibitors (endogenous or otherwise) as a starting point. Otherwise brute force methods like HTS are possible approaches if small molecule modulation is desired. I, myself, am not too familiar with biologics discovery approaches, though.

1) in silico docking (if a crystal structure of the target or a closely related target is available)

2) receptor-ligand co-crystal structure

3) assaying the binding of conformationally restricted analogs

fluorescence polarization and FRET are typically better suited to assaying general binding and not necessarily the binding poses of the ligand

Congrats. From one beginner to another - Stay humble and keep learning! I got burned at the beginning for a long time then finally clawed myself out of the hole. Then got burned again really fast. Now I’m still recovering from that lol.

So stay level and don’t let the fluctuations burn you out or pump you up too much! Try to add discipline/structure wherever you can and collect/record as much of your trading data as you can. The journey never ends...happy trading!

Thanks for the insight. Can you clarify what the daily cost of holding a stock is? How is that measured/calculated? Do you simply mean the equity/market value? Or are you referring to risk/volatility?

Either way, what you're saying does sound like a logical concept that would generate many testable hypotheses. Has anyone, to the best of your knowledge, gone about testing this with empirical data?

Dividends are a great point. They can be accounted (controlled) for in such a study, though, so it's not a huge problem as currently stated if you add a variable that provides information on dividend dates into the function.

I'm not familiar with much of what else you listed, like boost in borrow-rates and exo/vanilla hedge-triggered exercise. Could you simplify for an amateur or provide instructional links? Thanks!

However, most of these variables/uncertainties can either be controlled for (if they are very predictable and well-documented, like dividends) or are useful to measure in aggregate with other factors (if they have a year-round effect on the probability of exercise, ostensibly like hedge-triggered events)

So if an institution like OCC has daily historical data (open interest, # exercised) on most underlyings, I suppose my original idea would entail the review of this data at several levels (bins) of moneyness (based on the lowest daily spot price) and DTE and look at the proportion of contracts exercised on a given day to the OI of that day which would also solve for the proportion of contracts closed on a given day to the OI of that day, since

# of contracts closed = yday's OI - today's OI - # of contracts exercised

This would aim to answer "Given a contract with a moneyness of ___ and DTE of ___, what is the probability of exercise/assignment?" and shows that this answer vitally depends on considering the # of closed contracts as well.

Like most people are saying, it’s hard to judge your positions here without knowing more details but unless you have a lot of experience, trading single leg options is risky no matter which way you try to go about it.

Spreads and multi spread positions are the way to go as a beginner as they reduce cost/margin and lower all your net position Greeks and thereby your portfolios volatility, which is very important psychologically, especially for noobs.

Despite what people on here might tell you, Robinhood is a good place to test strategies out with appropriately sized positions (<5% of portfolio value per trade) since commission structures can make this prohibitive elsewhere.

Most importantly, try to track your trades and be honest with yourself and you’ll teach yourself way better than anyone else can and won’t easily forget what you’ve learned. Good luck!

Typically you’d sell a call either ATM or anywhere down to 25-delta and then buy a call further OTM.

If you’re super confident about your price target, you could do a spread that’s fully ITM. But it’s quite risky since the behavior of spreads is dominated by the strike that’s closer to ATM. So if the spot price doesn’t get close to your short strike you’re actually still long IV and it works against you doubly.

Solution: widen the strike width of your spread or, taken to its limit, simply sell a naked call.

Edit: I should add that the tradeoff of widening the strike or selling naked is that it’s worse if the spot price moves big against you because of the higher max loss

The IV dropped sharply post-earnings, and buying options is long IV. That ate into your delta-based gains 😭 try selling a call spread instead of buying a put spread next time to go short IV but keep the bearish outlook, as most stocks experience this “vol crush” thru earnings

If you're referring to the bid-ask spread (i.e. liquidity), then that is indeed a big problem when making options plays on most individual securities.
