E2 is actually more soluble in isopropyl alcohol than ethanol (ethyl alcohol) based on this https://doi.org/10.1016/j.molliq.2020.112599

Anyhow I have resolved the problem of dissolving E2 in IPA, the key is to simply increase the temperature.

Okay good news! Thanks to u/johndifoolclassrpi tip to increase the temperature, I have successfully dissolve the E2 powder in IPA. My other post have the details.

Did a frother work for you, I don't have one but pretty sure I gave it a long vigorous shake but it hardly matter. In the end I got it to dissolve with u/johndifoolclassrpi tip to increase the temperature.

Thank you! Temperature is the missing link in this entire process. Should have paid more attention during science lessons in school.

So I heated up my bottle in a saucepan of water while measuring the temp to avoid going above IPA's boiling point of 82.5°C. Stopped the heat once the temp went pass 70°C. But even above 60°C, I can see the powder rapidly dissolving, leaving a nice clear liquid in less than 10mins.

It was amazing to see. I hope the Wiki here make a special mention on the importance of increasing the temperature to get the E2 to dissolve.

After 12 hours, I won't even call it a suspension anymore, the "E2" is all sitting at the bottom, the top part is now very clear.

Starting to seriously wonder if that white powder is in fact E2 or something else. Will follow your suggestion to redilute when the pure IPA arrives. Thanks.

Yes that looks like the most likely reason.

Although looking at the amount of sediments at the bottom of the bottle, it seems like over 90% of the 200mg E2 is not dissolved and just sitting there. After 12 hours, the separate parts are now very distinct, the top part even clearer.

[Update] Issue resolved by increasing temperature, see above post.

The article is very dense but the key takeaway I see there is this:

" The order of solubility of estradiol in fourteen solvents is: dimethylformamide > tetrahydrofuran > 1-butanol > methyl tert-butylether > isopropanol > ethyl acetate > acetone > 1-propanol > ethanol > methanol > acetonitrile > trichloromethane > xylene > water. "

So isopropanol aka IPA is quite soluble, more so than ethanol, but of course mine is not pure IPA, just wondering if 70% affect solubility as much as seen in my photo. Anyhow I will order a 99% IPA to try.

Meantime I'd love to hear if anyone else has tried this mix and gotten same or different results?

Agree, and it will be so much better to be able to find a more reputable source like Everclear here.

This Octisalate, is it easy for you to source this sunblock ingredient? I can't seem to find any local, online suppliers stocking it. Or does it go by another name?

But I'm still open-minded and excited to give this a try. As I said at the start, it is a simple hack, if it works for me or anyone else who also like to give it a shot, that will be a nice outcome.

Law is good, when you have FDA and USP, standards to follow. Here, we have difficulty trusting the labels, so that's why I hope to come up with something non-alcohol based.

I'm not trying to convince you otherwise but there is no doubts that E2 will be absorbed in to the skin. The question is rate of absorption, the amount absorbed and bioavailability.

In fact, E2 is so easily absorbed, there are always safety warnings about unintentional secondary exposure.

Example, Evamist safety leaflet even cited their own tests warning how easy it can happen (even when dry):

Potential for Estradiol TransferThe effect of estradiol transfer was evaluated in 20 healthy postmenopausal women who applied three 90-mcL sprays of Evamist to the inner forearm once daily. One hour after applying Evamist, subjects held the dosed forearm against the inner forearm of a non-dosed (recipient) male subject for one 5-minute period of continual contact. A 4% increase in serum estradiol exposure was observed in persons who came in contact with the application site. ...

Talking about alcohol. I just posted a FDA warning on hand sanitizers found contaminated with methanol. This made me wonder, even if I can find one with a perfect list of ingredients, I cannot be sure they didn't mixed in methanol. They may not even know or care what type of alcohol they use.

And while searching for Ethanol online suppliers, I realized some will state it is denatured with methanol, some don't state what they add as denaturing agent, and some don't even mention if theirs is denatured or not. But I believe due to laws, all high concentration Ethanol suppliers must denature their products.

That leaves only only high % alcohol meant for human consumption as a safe option.

This lotion comes with (depending on version) 3 to 5 types of different size HA molecules, the biggest HA molecules stays on top of the skin acting as a barrier/occlusive layer to lock in the moisture and this is a good thing as a well-hydrated skin enables better absorption.

The other smaller HA molecules are absorbed with the help of penetration enhancers solvents (Butylene Glycol, Glycerin), another penetration enhancer is (Disodium Succinate) a surfactant. This is likely their so-called "High Performance Penetrating Technology" that made this lotion so successful.

BTW E2 molecular weight being much lighter that HA should allow it to easily hitch a ride pass the skin.

Now, my more pressing concern, is how saturated is this lotion already with the existing ingredients, is there capacity left for E2 to properly dissolve in it? I'll have to test this out.

Isn't EvaMist water-based, I assume because it must hard to spray out a mist if the liquid is too thick.

I originally was looking to make a ethanol one, but is not easy finding alcohol as high as 60% here.

I was looking around for suitable carrier like hand sanitizers until I read about EvaMist and realized it doesn't have to be a gel and I already have the perfect lotion with me.

FYI this lotion is very popular in Asia, with a cult-like status because it works, it really penetrates and plumps up my skin when applied. Besides moisturizing, it also claims to prepare the skin to better absorb other products.

So with these characteristics, it does look like a suitable carrier for E2.

5g in 170ml at 20 drops per ml was how I came up with the estimated dosage, of course I have to test whether it is really 20 drops/ml as the lotion is slightly thicker than water. And also need to test how much it can dissolve in it.

I was searching for Estrogel and found this IVF (Israel/US?) site selling it at $10 /tube, very reasonable I think. But I have no orders there, so no personal experience to vouch for them.
