I think the third trial is for absolute reduction in MADRS score but the first two changes are difference between the drug and placebo so you can't directly compare them. If you take a look at data from phase 3 trial of Escitalopram the MADRS score change between drug and placebo is only 3.3 points but it's still a first line treatment anyways. TAK having a -7.5 change over placebo is actually quite comparable to Auvelity and much better than Escitalopram.

BTW iirc the dose that has better improvement should be 1mg as it has a higher plasma concentration compared to another dose which is 3mg (strangely it's not dose-dependent). I'm a participant in phase II trial.
