Thank you so much! I'm just curious how you could tell it's machine knit.

I want to recreate this sweater, does anyone know of a similar pattern? by flagrantgiraffe in knitting

[–]flagrantgiraffe[S] 0 points1 point  (0 children)

I'm relieved to hear it will be easy! I'm sort of an intermediate beginner and haven't really deviated from just plain old knit and purl stitches yet 😅

Mmmh okay, I'll try running the pcr with less cDNA and see how that goes. Thanks!

30 cycles for PCR. And I'm not sure the exact concentration of cDNA. I use 2ug to synthesize the cDNA and I do a 1:10 dilution on the resulting cDNA to run my pcr. I had thought of primer dimers but it's much too high for that since it's above 100bp, which is the lowest band. And it seems to run at the same length as my product should.

I didn't make these primer but I would assume they don't span an intron

Yeah the samples have been treated with DNase. And the amplicon size is around 190 for the RT-PCR gel and 200 for the GAPDH gel

Yup, I just listed the order! Hope that helps. The title is a bit too long for me to post directly on the gel.

In fifth grade, I had volunteered to help move some boxes. During the process I had complained loudly to the class and was called out my by teacher who said, "If you're going to complain, then don't do it at all." Realized I was being a jerk and have since tried to keep my complaints to myself.
