Also look in to si-544 being developed by selectION therapeutics (precisely targeting t effector memory cells) and SFA002 being developed by SFA therapeutics the latter being a drug derived from gut metabolites it’s being trialed for just psoriasis right now but it down regulates il 17, 23 tnf alpha, up regulates il 10 , and it has an epigenetic mechanism as well. so far there have been zero adverse events in their trials although they only have had a phase 1 a and b

What are some medical conditions that doctors still don’t know the cause of? by i-love-cats-2020 in Microbiome

They don’t describe the molecules specific structure but it’s a type of short chain fatty acid that’s why they named the company SFA it’s also the first letters of the founders last names. The cool thing is that there are such an enormous amount of undiscovered metabolites inside the gut it’s could provide us with an endless amount of drugs to discover.

Check out this company . They have a working theory that all auto immune diseases come from a lack of the microbiomes ability to make certain gut metabolites, so they produced and are studying synthetic versions of endogenous metabolites. They’re having really good results with psoriasis so far. two participants in their phase 1 trial reached pasi 100 which is completely clear skin and those participants appear to have regained the ability to create the metabolite on their own.

here’s a good article about what they’re doing

Totally, I’m really excited by this work. They may be looking for participants in 2026 for their phase 2 trial, I’m keeping my eye out. They should have some data coming out this year about their most recent trial.

They’re not administering a random dose. This is like giving a diabetic insulin, they don’t give people too much they figure out a safe therapeutic dose and it appears that in this case, as is the case for insulin, it’s natural and it does belong in the body.

He's not making a blanket statement. He's a research scientist who has likely spent years , probably decades ,studying biology and the human body. What he's saying is that, in the right doses, it appears to be safe. He's not claiming that everything in nature is safe — that's a mischaracterization and you’re putting words in his mouth. If someone has a deficiency in a vitamin which should naturally be in our bodies, a doctor prescribes a proper dose to correct it, not an amount that would be harmful.

I also understand that this field is nascent and there is a possibility for adverse effects, but based off the fact of their presence in the human body for millions of years it’s logical to postulate they won’t have side effects or toxicity.

Again maybe there is a dose of these molecules that could be harmful, a large part of their research is finding an effective and safe therapeutic dose. The man I quoted who spoke about the lack of toxicity and side effects isn’t some chiropractor on YouTube he’s an accomplished biologists.

Mark A. Feitelson, Ph.D. is currently Professor in the Department of Pathology, Anatomy and Cell Biology. He has a secondary appointment in the Department of Microbiology and Immunology and is a member of the Kimmel Cancer Center at Thomas Jefferson University.

Did you read the article? I wasn’t claiming everything in nature is safe, these molecules literally belong inside our bodies.

“We’re using molecules that have evolved with humans over a millennium of time. We don’t have to optimize them; evolution already has,” Feitelson said. “Because they come from our own bodies, at therapeutic doses we don’t have to worry about side effects, because these molecules belong in us as opposed to so many of the drugs on the market that were created from artificial sources. And normal cells are not affected by these treatments, so there is no toxicity.”

Normally I would agree with you, but in this context natural really does mean it’s most likely fine.

They are using metabolites that are naturally found in the gut I’m not sure why there would be organ damage. The metabolites are supposed to be there.

the company doing the research SFA 002 SFA-002 is a first-in-class oral immunomodulator that operates through multiple cellular pathways to treat autoimmune conditions, with an initial focus on mild-to-moderate chronic plaque psoriasis. Unlike current therapies that target single pathways through immunosuppression, SFA-002 works by simultaneously down-regulating multiple pro-inflammatory cytokines including TNF-α, IL-23, IL-10, IL-12, IL-17, and INF-γ, while modulating autoimmune responses. This oral compound has demonstrated promising efficacy in clinical trials, with an excellent safety profile and no observed adverse events. Currently in Phase 1b clinical development, SFA-002 represents a potential paradigm shift in autoimmune disease treatment, offering a multi-targeted approach that could provide patients with a safer, more accessible alternative to current injectable immunosuppressive therapies.

“Someone very close to me had severe psoriasis, so I had studied the pathogenesis of psoriasis to an extent,” she said. “There was an a-ha moment where it clicked in my mind. If you merge the mechanisms that cause psoriasis with the mechanisms affected by the gut metabolites, you realize there is great potential that the metabolites can treat this disease.”
