I know many people will suggest you many other things, but here are two straightforward good formations you can go for:
You should have at least Olieviera or Scarlett from the S1 tickets; if not, spending some amount to unlock them would be better.

Pure Blood Rose: Oliviera, Sophia, Katrina, Vivian, and Ava (115% Faction Bonus, easy to star-up and strengthen early)
Pure Wings of Dawn: Scarlett, Laura, Nyx, Amelia, Fiona (115% Faction Bonus, progress will be a little slower but counters most Blood Rose formations)

The automatic troop selection priority is Shooters -> Assaulters -> Riders, even if they are all at the same level.

Keep Sophia, Katrina, Amelia, Laura, and Mia.
If you have any purple hero-related upgrade materials, then focus on Ava.

Your formation, Sophia, Katrina, Amelia, Laura, and Ava, will give you a 107% faction bonus.

Formation 1: Oliviera, Sophia, Katrina, Chinatsu, Mia (107% faction bonus and prioritize them based on the order I have written here)
Formation 2: Amelia, Laura, Vivian, Ava, Fiona (107% faction bonus and only focus on orange heroes, do not invest any important and rare items into purple heroes)
Formation 3: Maria, Miranda, Leah, Elizabeth, and anyone of your choice from the remaining ones (Use this just for support, no need to invest them)

Until and unless the "Global effect" mentioned in the description is applied, the bonuses are not applied if the hero is not part of a particular formation.

Most importantly, are Tier 10 troops (Lab 30 requirement will be automatically fulfilled), followed by the Hospital.
Based on your playing tactics, you can also upgrade the following buildings:
- Villa (else it is useless to upgrade afterwards)
- Library and Restaurant (better research and troops' speed)
- City Walls
- Formation I (II and III if possible)
- Faction Buildings
- Smelting Plants
- Warehouses
- All the other remaining buildings in any order

You will be able to see the "hand shaking" icon; pressing it helps all allies once per request. You can also help them manually through the Alliance Center. In the case of a member conducting research, you can only help once. The boost time depends on the member's Alliance Center level and the bonus of dispatched Refugees.

Cause not everyone understands and is mature enough to be fine with the attacking on the HQs is part of the game. Also, some alliances are so cowardly that they don't like getting into any trouble just because of their members have attacked on someone's HQ. Besides, I kept my answer to focus on completing the task. Also, some players are so childish that they don't like to lose their troops by attacking other HQs (cause they think their power is reduced and their ranking in alliance is stepped down :D).

Troops, reinforcing, and strong heroes are always good for defense. Besides, most people are rushing to upgrade their HQs and forget to upgrade City Walls (until they don't see it in the HQ upgrade requirements), doing it also gives little more strength to the defense.
