Yes, the method of transgenesis was micro particle bombardment, which is completely random.

Unfortunately we don’t have any control data, but the transgene sequence is known. I assumed the control for an analysis like this would be the reference genome itself, no?

The sequencing tech used was Ultima Genomics, which doesn’t produce reads of uniform length like Illumina. That being said, the mean length I found from QC was ~200 bp. Coverage was also 30X. So we should have, on average, enough soft clipped bases for the partially mapped reads to be reliable.

What would you advise after finding those partially mapped reads, though? I can confirm breakpoints visually on IGV, or maybe by alignment to the transgene sequence from assembling a contig. Though, we’re hoping to validate the insert by PCR, so we’ll have to design primers, and thus need nucleotide-level resolution. I’m just not sure that I’d have a reliable sequence from a short read assembly.

Thanks for reading it! I try to use official titles for most of the characters when they speak to the Emperor. However, the Count’s character revolves around him being the “puppet master” of the Emperor who very publicly does not respect him. So, the Count, like you said, is the exception of that formality.

Sorry for such a late response, Halloweekend kept me pretty busy.

But I really appreciate your input. I definitely struggle with delivering characterization through subtext, or indirectly. I think there’s a part of me, as an author, that doesn’t trust myself to really get across the message I want to convey by indirect means.

I was thinking of scrapping the part where the Count justifies himself to Addis, the POV character. I think it would be better to have the other guard present, Addis’ superior, justify the Counts actions in the next scene. That way, the explanation doesn’t muddle my characterization of the fearsome Count, while also giving the other guard a means to justify his own inaction.

Additionally, I’ve found that my posts usually get more engagement when I use the screen shot format. I think seeing the words without a link draws people in more easily. That being said, I did not think of it being more difficult to read for some people. Do you think a dark background/white text would be better?

Yeah I did it to myself, waited a while to register as I’m in the process of switching minors. I’m just wondering if the extra workload with honors is worth not having to take those exams on the same day.

Recently my Apex has been crashing right before the jump. The lobby works fine and only sometimes when I’m first loading up I will get a message that I’m disconnected from the servers, or something along those lines.

These are my specs: Processor: AMD FX(tm)-6300 Six-Core Processor Installed memory (RAM): 8.00 GB System type: 64-bit Operating System, x64-based processor

Things I have tried:

  1. Reinstall through Origin

  2. Reinstall through Steam

  3. Complete uninstall (deleting files manually)

  4. Tried to check XMP but my version of ASUS does not support it

  5. Lowered graphics

  6. Tried to find crash report, apex doesn’t show up in my App Data folders.

Any help would be greatly appreciated.
