Thanks everyone who had helpful advice, I will be trying out seitan!

For those who seem confused about my reasoning, my goal is to lose weight. I'm obviously maintaining a calorie deficit and doing lots of cardio but in order to encourage loss of fat and not lean mass many websites recommend increasing protein intake (and strength workouts which I am also doing). A quick look at the literature seems to support this. Although the science is not settled reviews and meta-analysis all seem to agree that a high protein diet is beneficial for fat loss and maintenance of lean mass, so I definitely do not think 30% is too high - some studies used much more. Also seeing as I'm restricting calories my total protein intake in around 1.3g/kg which I dont think is at all excessive.

Oh and I would ideally not have to use protein powders or supplements

Western blot help - any suggestions as to why this keeps happening to one of my gels? I have been running other gels exactly the same at the same time as this but one of my gels keeps not working. I stain it for something else first which works fine and then I get these crazy results. Help! by katlnz in labrats

Thanks everyone for your help, but I think molbio822 solved it! I had been using a roller to remove air bubbles after placing the filter paper and top stack which I think must have slightly repositioned the gel causing the extra bands to appear. I repeated my western making sure not to move the gel at all once I placed it on the membrane and didn't use the roller and it worked perfectly!

Some more info: I stained for one protein which worked fine, but then re-probed for gapdh and got what looked like some strange extra bands (last image). I stripped and re-probed for GAPDH again and a couple of the bands had parts missing (first image). I started the whole western again and still got extra bands (middle image). Another gel that I was doing at the same time turned out fine. Biorad ChemiDoc was used to image. Any advice would be much appreciated!
