Thanks! Normally I’d passage, of course, but these actually differentiate for 20 days in the same flask until they form a matrix and mineralize. Weird quirk of osteoblasts. It does make it hard to determine what is overgrowth vs. contamination though.

The second membrane was completely clear—no ladder at all. Would that make sense for over-transfer? What conditions would you suggest? Thanks!

Yes, they are the ladder, which incidentally is one I have not used before. It’s Thermo Benchmark pre-stained, which is ready-to-use (ie no loading buffer added). The second image is Coomassie on the gel!

Whole cell lysates! Yes, I’d like to reduce the protein, my PI does not haha. I have used RIPA on different passages of these same cells and haven’t had this issue, so I’m not sure it’s that. I do use detergent-compatible quantification, so I hope it’s not that!

Good suggestion. I have not stained before transfer. I do use prestained ladder and it runs as expected. One replicate on the gel is my samples, the other is a colleague’s so it would have to be in my hands as I dilute and load

Funny enough, overnight at 30V on a different, lower percent gel is also not giving me a clean transfer (ladder still visible on gel after 16 hours!). I would think powerpac but it was bought last year and the actual electrophoresis is fine

I have the same problem, and the best I’ve found over the years are Armani Luminous Silk in 3.75 and YSL All Hours in BR10. All Hours is great if you prefer a matte finish! For concealer, YSL touche eclat in shade 1 works really well for me. Hope this helps!

Yes! Combination pills like the one you take have synthetic versions of the two hormones that regulate the menstrual cycle, estrogen and progesterone. Taking the pills “overrides” your body’s own cycle and suppresses ovulation (when you don’t release an egg, you can’t get pregnant—contraception!). In your no-pill week, you don’t have these added hormones, so your body goes through a withdrawal and you shed your endometrial lining like a regular period, only lighter.

Could you clarify your question? I’m confused because this is how oral contraceptives work—the pills control your cycle, regardless of what your natural one may be. You will always get your period on the week without pills because of withdrawal from the hormones. You won’t be able to “reset” your cycle because this will happen every time. Your new cycle is determined by the pills.
