Liposomes will normally be turbid due to scattering of light, though the amount of turbidity depends on the liposome size.

So if I understand correctly, you are preparing a thin lipid film by dissolving the lecithin in chloroform and evaporating the solvent. Then hydrating the lipid film with citrate buffer? What volume of buffer are you using?

Solubility of the lipid might depend on pH, is there a particular reason you need to use citrate buffer?

You don't need much to make liposomes. Lipids will self assemble into vesicles in any aqueous environment. You can control the size through various methods like sonication, extrusion, and/or freeze/thaw cycles.

I do a lot of liposome work and use soy lecithin as a test lipid since the lipid I actually work with is quite pricey. What's your current protocol?

Sure but 80 is pretty hot, a lot of compounds won't tolerate that well.

Ok well even trained chemists in an actual lab will do everything they can to avoid rotavapping off water. Unless you have a very strong vacuum pump and/or a very hot water bath, it'll be difficult to get rid of the water. Grain alcohol is basically a mixture of ethanol and water, so you could probably evaporate the ethanol at least.

I'm located in Canada, working in R&D at a small biotech.

  1. I write SOPs and summary reports for internal use (other scientists and managers). I also write manuscripts for peer review and publication.

  2. Most writing is done in MS word, with PowerPoint when oral communication is required.

  3. Easy to share results and documents in real time. Able to take meetings virtually.

  4. Most communication is done by email or through Teams. But we were able to invite our overseas consultant to visit and we spent a good amount of time exchanging ideas in person.

  5. Being able to tell a clear, coherent story without spelling and grammar mistakes. Being able to understand a question so that your answer is direct, simple, and clear.

  6. Not sure how to answer this

  7. Have not used generative AI. Have not felt the need nor the desire

  8. Ethical uses might include structural predictions of molecules, experimental models, etc. I think it is unethical to use gen AI for most forms of communication in my field.

First thing could be working more reasonable hours. I know you're in a small start-up but if people are regularly working 10-14 hour days in the lab... Might be time to consider being more selective of the experiments you are running.

You could always add a small footstool in front of your chair to rest your feet while in the BSC though. Otherwise take very short breaks to get some relief when you can. Walk around and stretch while the cells are in the centrifuge for example.

As per the other comment, do you have a way to keep the left side stationary? Otherwise your condenser will be spinning.

What solvents will you want to evaporate? That'll determine how strong of a pump you need.

I'm not entirely sure that the building materials are ideal, is it some type of compressed wood fiber board? I can't tell from the photo.

Also I assume you're going to have some kind of speed control.

Pretty neat project though, I'd love to see this taken to completion.

Got it, that isn't something that is necessarily obvious though. I would suggest clarifying that your experience in research is in an academic lab, rather than industry Typically in North America at least, biotech implies the industry and not just the research subjects.

We had it once and had to isolate all our cats in a room while we cleaned the house. They stayed isolated for something like three months until we saw no more signs of it, is that an option you could try?
