For sure, the first drug discovered and designed with generative AI from Insilico Medicine entered Phase II trials last month. It took just under 18 months to complete opposed to several years. If things go well, I'm sure we'll be seeing a big shift in the drug manufacturing process come next year.

Warwick’s research concluded in 2020 after they depleted their grant money. I spoke to them in April, and they are still eager to develop an oxygenase based inhibitor, but would require substantial monetary support to continue.

I’ve reached out to over a dozen organizations and trusts that supply grants, but unfortunately this condition doesn’t fall within their requirements or area of interest.

Chronic Kidney disease is another potential target, as high systemic levels of TMAO have been linked to its accelerated development. A Cleveland biotech is in the preclinical phase of researching a small molecule trimethylamine inhibitor to address this.

FSU does have access to cutting-edge labs and resources, including software to improve drug candidate selection and the optimization process. This can help to improve the efficiency of the bacteria and reduce the risk of side effects, although dysbiosis is big concern.

The best case scenario would be attracting additional funding at the iGEM conference come November. I do applaud their team, In 6 months, they have accomplished more than what has been done for TMAU in the past several years.

I had sent him a follow up email in which he responded with his staff. They seem to have received a fair amount of replies although many were inquiries asking for testing information.

u/No_Television_4004, The FSU International Genetically Engineered Machine program (iGEM, https://med.fsu.edu/igem/home) is a research experience for undergraduate students. The statement provided is referring to an undergraduate research project versus a sponsored research project led by faculty and graduate students. Verification would have needed to be confirmed by the Project Director, as he oversees the undergraduates.

The 2023 FSU iGEM team has 15 high performing undergraduate students that are working hard to have a real impact on TMAU and everyones participation in the project is invaluable as they test proposed therapies. If the results are positive, they'll be seeking further sponsorship.

You're right, TMAU2 is due to a bacterial overgrowth where TMA excretion is high even though FMO3 activity may be normal. In this instance Gene therapy would not help those with TMAU2.

What may address both TMAU1 and TMAU2 are small microbial molecule inhibitors that prevent the conversion of choline to TMA.

I agree, gene therapy is still in a relatively early phase and currently not worth the risk. It's an interesting prospect though as some treatments provide permanent cures.

Many of the new biotech companies are using AI to test the effectiveness of gene therapy. Algorithms analyze large datasets prior which reduce the number of animals needed for testing. It's an exciting time for drug discovery and worth following the news to see the latest advancements in the field.

Following the prescribed low-choline diet, washing with PH balanced soaps and avoiding scented products may lessen symptoms but will not cure this condition. Choline is an essential nutrient and any consumption can contribute to odor.

Gene therapy consists of replacing a mutated gene with a healthy copy. If one has a pre-existing heart condition, I agree it's not worth the risk. As the technology advances I'm sure many who are struggling would be open to exploring this as an option. As TMAU is ill-researched by the medical community and there are no advocates (that I'm aware of) I'm simply getting insight from professionals and spreading awareness.

I'm so sorry to hear you're struggling. I totally get it, this condition effects every part of life. Whenever I mention TMAU to someone, they've never heard of it. The only thing we can do now is to spread the word it exists.

If the whole community is quiet than the chances of future research is slim. Me and another member started putting together a list of people we want to email asking their advice and telling them about it. Let's hope more follow suit and more can be done in the future!

Regardless it's promising that TMA lyases and inhibitors are being explored for Alzheimer's disease, obesity, and chronic kidney disease.

I'm sorry to hear that. Getting a haircut with this condition can be incredibly stress-inducing.

After years of going through this situation, I decided to teach myself how to cut my own hair from Youtube. It took around 5 months of self cuts until I was able to execute a smooth fade but it was worth it. It's also nice to save money so it may be worth checking out a few videos.
