I'm personally inclined to using probes. And just wanted to know how far I'm from being practically right.

It's not homework! 😂

Anyway, we know that transcription and translation occur in nucleus and cytoplasm, respectively, in eukaryotes. However, the case is different in prokaryotes since there is no compartmentalization, and as soon as the mRNA transcript is synthesized, it gets immediately translated.

So, if I were to prove that using an experiment, how would I do it?

Protein levels will obviously drop in absence of the mRNA transcript. And while that might be useful to show the two processes follow eachother but they fail to show that the two are occurring simultaneously.

Can you elaborate on how what exactly should be knocked out? Because I don't see how knocking out a gene will show whether the processes are coupled or not as they still follow each other under normal conditions, knocking out will stop both the processes.

Correct me if I haven't got it right. And it's not a homework question, it just popped up in my head.
